<dcterms:abstract xml:lang="fr">Les mitochondries sont des organites possédant leur propre génome qui peut être l’objet de mutations, souvent associées à de graves maladies neuromusculaires. La plupart des mutations de l’ADN mitochondrial (ADNmt) sont hétéroplasmiques, ce qui signifie que les molécules mutantes et sauvages coexistent dans une même cellule. Ce projet de thèse a été consacré au développement de deux stratégies qui visent à diminuer la proportion d’ADNmt mutant en utilisant les voies d’import mitochondrial d’ARN. Nous avons tout d’abord utilisé une stratégie anti-réplicative qui vise à altérer la réplication de l’ADNmt mutant. Nous démontrons que l’expression d’ARNr 5S contenant des séquences anti-réplicatives permet de diminuer le taux d’hétéroplasmie dans des conditions sélectives de culture. Puis, nous avons développé un système CRISPR/Cas9 mitochondrial pour cliver l’ADNmt. La nucléase Cas9 et des ARNsg modifiés peuvent être redirigés vers les mitochondries. L’introduction de deux ARNsg ciblant des régions distinctes de l’ADNmt provoque des déplétions de ce génome, ce qui pourrait constituer la première indication d’un effet de CRISPR/Cas9 sur l’ADNmt.</dcterms:abstract>
<dcterms:abstract xml:lang="en">Mitochondria are organelles containing their own genome which can be affected by mutations often associated with severe neuromuscular disorders. Most of the mitochondrial DNA (mtDNA) mutations are heteroplasmic, meaning that mutant and wild-type molecules coexist in a same cell. The objective of this thesis project consists in the development of two strategies to decrease mutant mtDNA proportion by use of RNA mitochondrial import pathway. We first applied an anti-replicative strategy aiming to alter mutant mtDNA replication. We demonstrated that the expression of 5S rRNA containing anti-replicative sequences allows to decrease the heteroplasmy level in selective culture conditions. Then, we developed a mitochondrial CRISPR/Cas9 system to cleave mtDNA. Modified Cas9 nuclease and sgRNAs can be imported into mitochondria. Delivery of two sgRNAs targeting distinct regions of the mitochondrial genome induced mtDNA depletion, thus providing a first indication of a CRISPR/Cas9 effect on mtDNA.</dcterms:abstract>
