<dcterms:abstract xml:lang="fr">Ce travail de thèse est focalisé sur le développement de méthodes combinant la chromatographie liquide non dénaturante à la spectrométrie de masse native (MS native) pour la caractérisation de protéines thérapeutiques en particulier des anticorps monoclonaux (mAbs). Les techniques de chromatographies liquides non-dénaturantes (ndLC) sont utilisées avec des phases mobiles comportant des sels non-volatils et donc incompatibles avec la MS native. Dans ce contexte, les développements de nouveaux couplages impliquant la chromatographie d’exclusion stérique (SEC), la chromatographie à échange cationique (CEX), la chromatographie d’interaction hydrophobe (HIC) et la MS native ont été réalisés. Le couplage de la SEC à la MS native a permis d’identifier et de quantifier les variants de tailles de mAbs. Le couplage CEX-MS native permet l’identification et la quantification des variants de charges de mAbs. Enfin, le couplage direct de la HIC à la MS native a permis de caractériser les anticorps immunoconjugués à cystéine. L’ensemble des développements méthodologiques réalisés au cours de ce travail de thèse ont permis de démontrer le bénéfice des couplages ndLC-MS native.</dcterms:abstract>
<dcterms:abstract xml:lang="en">This thesis work focuses on the development of analytical methods hyphenating the non-denaturing liquid chromatography and native mass spectrometry (native MS) for therapeutic protein characterization, especially for monoclonal antibodies (mAbs). Non-denaturing liquid chromatography (ndLC) techniques are performed by using mobile phases composed of non-volatile salts and inherently not compatible with native MS. In that context, the developments of new couplings involving size exclusion chromatography (SEC), cation exchange chromatography (CEX), hydrophobic interaction chromatography (HIC) and native MS were realized. The hyphenation of SEC to native MS allowed the identification and quantification of mAb size variants. Moreover, CEX hyphenated to native MS provided an identification and quantification of mAb charge variants. Finally, the hyphenation between HIC and native MS allowed the characterization of cysteine-linked antibody-drug conjugates. Consequently, the benefit of non-denaturing liquid chromatography hyphenated to native MS was outlined throughout this thesis work.</dcterms:abstract>
