<dcterms:abstract xml:lang="fr">Au cours de ce travail, nous avons étudié le crosstalk fonctionnel entre récepteurs opioïdes mu et delta en conditions récompensantes induites par des stimuli naturels tels que la nourriture appétente et/ou l’exercice physique ou par des drogues telles que la morphine. En utilisant les souris doubles knock-in mu-mCherry /delta-eGFP, nous avons montré que l’hétéromérisation de mu et delta altère la signalisation et le trafic du récepteur mu en réponse au peptide opioïde met- enképhaline, mais pas β -endorphine, en culture primaire d’hippocampe et in vivo. Nous avons également montré qu’une administration chronique de morphine élargit la coexpression neuronale de mu et delta dans le cerveau. Ces changements persistent après 4 semaines d’abstinence indiquant leur caractère durable. Dans la seconde partie de la thèse, nous avons cartographié les connexions de la région subfornicale de l’hypothalamus latéral (LHsf) chez la souris et nous avons examiné l’activation des récepteurs mu et delta dans cette région après privation de nourriture et restauration de l’accès à des aliments riches en gras ou contrôle. Dans le LHsf, connecté de façon réciproque à de nombreuses régions de l’hypothalamus ou en lien avec le système de la récompense, la privation de nourriture induit l’internalisation des récepteurs delta quel que soit le régime alimentaire tandis que l’accès à la nourriture active les récepteurs mu et delta différemment selon le type de régime alimentaire. Enfin, l’expression des gènes opioïdes a été mesurée chez des rats nourris de façon chronique avec un accès libre au gras et au sucré et la possibilité de faire de l’exercice physique. Une interaction entre ces deux paramètres n’a été mise en évidence que sur l’expression du récepteur delta dans l’hypothalamus latéral.</dcterms:abstract>
<dcterms:abstract xml:lang="en">In the thesis, we studied the functional cross-talk between mu and delta opioid receptors in rewarding conditions elicited by natural stimuli such as palatable food and/or exercise or by drugs of abuse such as morphine. Using mu-mCherry/delta-eGFP double knock-in mice, we showed that mu-delta heteromerization alters mu opioid receptor signaling and trafficking in response to the endogenous opioid peptide met-enkephalin, but not β-endorphin, in primary hippocampal cultures and in vivo. We also showed that chronic morphine administration extended mu-delta neuronal co-expression throughout the brain which persisted after 4 weeks abstinence, pointing to morphine-induced long- lasting changes. In the second part of the thesis, neuroanatomical connections of the subfornical area of the lateral hypothalamus (LHsf) were mapped in mice and examined the activation of mu and delta receptors in this region following fasting and refeeding in HF diet and chow diet. Within the LHsf, which was reciprocally connected to many hypothalamic and reward related brain areas, fasting internalized delta receptors irrespective to the diet regimen whereas refeeding differentially activated mu and receptors in chow-fed and HFD-fed animals. Finally, opioid system related gene expression was measured in the long-term fc-HFHS fed and voluntary wheel running rats, which only revealed an interaction effect for delta opioid receptor expression in the LH.</dcterms:abstract>
