<dcterms:abstract xml:lang="fr">Il existe deux types d’ARN : les ARN messagers qui sont des ARN codants, et les ARN non codants.La machinerie responsable de la dégradation des ARN qui ne sont plus nécessaires ou qui sont endommagés contribue au contrôle de l’expression des gènes chez tous les organismes vivants. Les ribonucléases sont les enzymes catalytiques de cette machinerie. Leur caractérisation est donc cruciale pour comprendre la manière dont les cellules adaptent leurs programmes d’expression génétique en fonction des signaux extracellulaires. Dans cette thèse, une nouvelle ribonucléase humaine appelée HELZ2 a été identifiée et caractérisée. Ses capacités à dégrader l’ARN ainsi qu’à hydrolyser l’ATP in vitro a été démontrée. Les caractéristiques de son domaine nucléase appelé RNB ont été décrites. Sa localisation, majoritairement cytoplasmique a aussi été démontrée. Cette étude lie clairement la protéine HELZ2 au métabolisme des ARN permettant d’étudier ses fonctions in vivo sous un nouvel angle.</dcterms:abstract>
<dcterms:abstract xml:lang="en">Two types of RNAs are known: messenger RNAs that are coding RNAs, and non-coding RNAs. The machinery of RNA decay which degrades damaged RNAs and those that are no longer needed contributes to the control of gene expression in all living organisms. Ribonucleases are the catalytic enzymes of this machinery. Their characterization is therefore crucial to understand how cells adapt their gene expression programs to extracellular signals. In this thesis, a new human ribonuclease called HELZ2 has been identified and characterized. Its ability to degrade RNA as well as to hydrolyze ATP in vitro has been demonstrated. The characteristics of its nuclease domain called RNB have been described. Its predominantly cytoplasmic localization has also been demonstrated. This study clearly links HELZ2 protein to the RNA metabolism providing a new angle to study its functions in vivo.</dcterms:abstract>
