<dcterms:abstract xml:lang="fr">Les micro (mi)ARN sont des régulateurs post-transcriptionnels de l’expression génique. Retrouvés chez la plupart des eucaryotes, ils sont impliqués dans un grand nombre de processus biologiques. Certains virus sont capables de détourner cette voie à leur avantage et expriment leurs propres miARN ou modulent l’expression de miARN cellulaires. Ce projet porte sur la caractérisation du rôle joué par la protéine effectrice Argonaute (AGO) 2 dans l’établissement de l’infection par le cytomegalovirus murin (MCMV). Outre l’expression de miARN viraux, celui-ci est capable d’induire le phénomène de dégradation du miARN cellulaire miR-27. L’importance d’AGO2 dans l’accumulation des miARN cellulaires et viraux, ainsi que la recherche de ses partenaires protéiques, ont été déterminées au cours de l’infection par le MCMV.</dcterms:abstract>
<dcterms:abstract xml:lang="en">Micro (mi)RNA are post-transcriptional regulators of gene expression. Found in most eukaryotes, they are involved in a wide range of biological processes. Some viruses are able to hijack this pathway for their benefit and express their own set of miRNA or regulates cellular miRNA expression. This project is focused on the characterization of the role played by the effector protein Argonaute (AGO) 2 in establishing murid cytomegalovirus (MCMV) infection. Additionally to viral miRNA expression, this virus is able to induce the phenomenon of target RNA-mediated miRNA degradation of miR-27. The key role of AGO2 in cellular and viral miRNA accumulation and the identification of his protein partners had been determined during MCMV infection.</dcterms:abstract>
