<dcterms:abstract xml:lang="fr">Le paludisme est une maladie mortelle causée par le parasite Plasmodium falciparum. L'artémisinine (Art) est le médicament antipaludique le plus efficace, mais est menacée par l'émergence de la résistance à l'Art (Art-R). Les jeunes anneaux et les gamétocytes matures sont respectivement impliqués dans l'Art-R et la transmission du parasite aux moustiques anophèles. Il est donc urgent de trouver de nouveaux agents qui peuvent contourner Art-R et bloquer la transmission. La Plasmodione (PD) a déjà révélé son efficacité sur les anneaux et une synergie avec l'Art. En premier, les travaux de doctorat ont démontré que la PD présente une résistance croisée avec l'Art dans les stades asexués de P. falciparum Art-R. Ensuite, afin d'identifier l'interactome de la PD chez les parasites, des sondes photoréactives et clickables ont été évaluées/validées par une analyse détaillée des adduits covalents générés sous photoirradiation et réaction click en présence de différents partenaires de réaction.</dcterms:abstract>
<dcterms:abstract xml:lang="en">Malaria is a fatal disease caused by the parasite Plasmodium falciparum. Artemisinin (Art) is the most effective antimalarial drug, but is threatened by the emergence of resistance to Art (Art-R). Young rings and mature gametocytes are involved in Art-R and transmission of the parasite to Anopheles mosquitoes, respectively. Therefore, it is urgent to find new agents that can bypass Art-R and block the transmission. Plasmodione (PD) was previously shown to be active on rings and to display synergy with Art. First, the PhD work demonstrated that PD or its analogue bMD40 displays cross-resistance with Art in asexual stages of Art-R P. falciparum strains. Secondly, in order to identify the PD interactome in the parasites, ‘clickable’ and photoreactive activity-based protein profiling probes were evaluated and validated by detailed analysis of covalent adducts generated upon photoirradiation and click reaction in the presence of different reaction partners.</dcterms:abstract>
