<dcterms:abstract xml:lang="fr">Les stratégies de protéomique quantitative sont très attractives pour l’étude des systèmes biologiques au niveau protéique. Elles peuvent être scindées en trois grandes catégories : les stratégies globales, les stratégies ciblées et les stratégies DIA qui combinent les avantages des deux premières. Ce travail de thèse est axé sur le développement de stratégies analytiques et bioinformatiques en protéomique quantitative, afin d’améliorer la quantification de protéines présentes à l’état de traces. La première partie de mon travail a consisté à optimiser différentes étapes du protocole analytique en me concentrant tout particulièrement sur le mode d’acquisition DIA et le traitement de ces données complexes. Par la suite, différentes méthodes de quantification ont été éprouvées dans plusieurs contextes biologiques afin de répondre au mieux aux problématiques posées. Enfin, différentes stratégies analytiques ont été développées pour étudier les interactions protéine-protéine, afin de mieux appréhender certains processus biologiques, à savoir la biogénèse du ribosome et la régulation des ARNm.</dcterms:abstract>
<dcterms:abstract xml:lang="en">Quantitative proteomics strategies are very attractive for the study of biological systems at the protein level. They can be divided into three main categories: global strategies, targeted strategies and DIA strategies that combine the advantages of both. This PhD work is focused on the development of analytical and bioinformatics strategies in quantitative proteomics for improving the quantification of trace-level proteins. My first focus consisted in optimizing different steps of the analytical protocol by focusing on the DIA acquisition mode and the processing of these complex data. Subsequently, different quantification methods have been evaluated in several biological contexts to properly address the issues involved. Finally, different analytical strategies have been developed to study protein-protein interactions to better understand various biological processes, namely ribosome biogenesis and mRNA regulation.</dcterms:abstract>
