<dcterms:abstract xml:lang="fr">La détection de biomarqueurs cancéreux pour le diagnostic est un enjeu clé dans la prise en charge et le traitement de la maladie. Le but du projet est de développer des méthodologies simples et rapides pour la détection d’ARN impliqués dans le cancer dans des échantillons cellulaires, à l’aide de nanoparticules (NPs) de polymères fluorescentes fonctionnalisées d’ADN. Avec des nanosondes basées sur le transfert d’énergie (FRET), nous avons développé une technique de détection de micro-ARN dans les lysats cellulaires. Nous avons aussi mis au point une méthode de détection d’ARNm dans les cellules fixées, beaucoup plus rapide et sensible que la méthode FISH classique. Enfin pour délivrer les nanosondes dans les cellules vivantes, l’étude systématique d’électroporation de NPs non-fonctionnalisées a été réalisée. Les méthodologies développées à base des NPs constituent des outils robustes pour la détection et l’imagerie de biomarqueurs cancéreux pour la recherche et le diagnostic médical.</dcterms:abstract>
<dcterms:abstract xml:lang="en">The detection of cancer biomarkers in medical diagnostics is crucial for early detection and treatment of the disease. The aim of the project is to develop simple and rapid methodologies for the detection of RNA biomarkers of cancer in cell samples, using DNA-functionalized fluorescent dye-loaded polymer nanoparticles (NPs). Using nanoprobes based on energy transfer (FRET), we have developed a technique for the detection of microRNA in cell lysates. We have also introduced a method for mRNA detection in fixed cells, which is much faster and more sensitive than the classical FISH method. Finally, to deliver the nanoprobes into living cells, the systematic study on electroporation of non-functionalized NPs was carried out. Overall, based on fluorescent nanoprobes, we developed robust methodologies for the detection and imaging of cancer biomarkers in research and medical diagnostics.</dcterms:abstract>
