<dcterms:abstract xml:lang="fr">La métabolomique par spectroscopie RMN HRMAS permet de travailler directement sur des tissus biologiques hétérogènes intacts ; elle identifie et quantifie de nombreux métabolites avec une excellente sensibilité. Nous l’avons utilisée pour étudier l’hétérogénéité du métabolisme des gliomes et établir un lien avec la biologie moléculaire et le pronostic. Différents biomarqueurs pronostiques ont ainsi été identifiés. Ces biomarqueurs montrent les voies métaboliques privilégiées utilisées par les gliomes et pourront devenir de nouvelles cibles thérapeutiques. Par ailleurs, nous avons mis au point des outils d’analyse des voies métaboliques en utilisant des sondes moléculaires marquées au 13C sur cultures cellulaires et modèles de xénogreffe dans la perspective d’une application chez l’Homme.</dcterms:abstract>
<dcterms:abstract xml:lang="en">Metabolomics by HRMAS NMR spectroscopy allows working directly on intact heterogeneous biological tissues; it identifies and quantifies many metabolites with an excellent sensitivity. We have used this method to study the heterogeneity of glioma metabolism and related it to molecular biology and tumoral prognosis. Different prognostic biomarkers have thus been identified. These biomarkers show the preferred metabolic pathways used by gliomas and may become new therapeutic targets. In addition, we have developed tools for the analysis of metabolic pathways using 13C-labelled molecular probes on cell cultures and xenograft models with a view to their application in medicine.</dcterms:abstract>
