<dcterms:abstract xml:lang="fr">Les mécanismes de dégradation des ARN ont un rôle majeur afin d’assurer la régulation de l’expression des gènes. Ces phénomènes sont cruciaux lors des transitions développementales et en conditions de stress biotiques et abiotiques. Au cours de ma thèse j’ai caractérisé une nouvelle endoribonucléase à domaine NYN, DAE (DCP1 Associated Endoribonuclease). J’ai montré que DAE interagit directement avec l’activateur de decapping DCP1. L’analyse du transcriptome de plantes dans lesquelles l’expression de DAE est altérée suggère qu’elle agit en parallèle du complexe de decapping. Mes résultats révèlent l’existence d’un nouveau complexe, distinct du complexe de decapping canonique et dont DAE est la sous-unité catalytique. L’étude de la fonction biologique de cette nouvelle voie de dégradation des ARNm permettra de mieux comprendre comment ces mécanismes sont orchestrés chez les plantes afin de répondre à des modifications de leur environnement.</dcterms:abstract>
<dcterms:abstract xml:lang="en">RNA degradation pathways have a major role in the regulation of gene expression. These mechanisms are crucial determinants of developmental transitions and in the plant response to biotic and abiotic stresses. During my PhD, I characterized a hitherto unknown NYN domain endoribonuclease, DAE (DCP1 Associated Endoribonuclease). I established that DAE interacts directly with the decapping activator DCP1. The transcriptomic analysis of plants with altered DAE expression, strongly suggest that it acts in parallel of the decapping complex. Overall my results reveal the existence of a new complex distinct from the canonical decapping complex, in which DAE is the catalytic subunit. Studying the biological function of this new complex will improve our understanding of how RNA degradation pathways in plants are orchestrated to respond to environmental stimuli.</dcterms:abstract>
