Les cancers HER2+ sont caractérisés par l’amplification et la surexpression du récepteur tyrosine kinase HER2. STARD3 est un gène co-amplifié et co-exprimé avec HER2 dans les cancers. STARD3 est une protéine endosomale de la famille des START qui lie le cholestérol et qui interagit avec les protéines VAPs ou MOSPD2 pour créer un site de contact membranaire entre les endosomes tardifs et le RE. Au niveau de ces contacts, STARD3 transporte le cholestérol du RE vers les endosomes aux dépends de la membrane plasmique (MP). La perte d’expression de STARD3 inhibe spécifiquement la croissance des cellules HER2+. STARD3 agit sur la croissance des cellules HER2+ de manière indirecte en altérant la distribution normale du cholestérol et en modifiant la composition lipidique de la MP. Ces modifications favorisent la dimérisation d’HER2, son activation et l’activation de la voie PI3K/Akt. Ainsi, les cellules HER2+ dépendent du transport de cholestérol médié par STARD3 pour leur croissance.
HER2+ cancers are characterized by amplification and overexpression of the tyrosine kinase receptor HER2. STARD3 is a gene co-amplified and co-expressed with HER2 in cancers. STARD3 is a late endosomal cholesterol-binding protein that belongs to the START protein family. STARD3 interacts with ER proteins (VAPs or MOSPD2) to create membrane contact sites between LE and the ER. At these contacts, STARD3 transfers cholesterol from the ER to LE inducing a cholesterol accumulation in LE at the expense of the plasma membrane (PM). Inhibition of STARD3 expression inhibits specifically HER2+ cancer cell growth. STARD3 acts on HER2+ cancer cell growth through an indirect mechanism by altering the normal cholesterol distribution and modifying the lipid composition of the PM. These changes promote the dimerization and activation of HER2 and the activation of the PI3K/Akt pathway. Indeed, HER2+ cell growth depends on STARD3-mediated cholesterol transport.
