<dcterms:abstract xml:lang="fr">Les aminoacyl-ARNt (aa-ARNt) sont essentiels à la synthèse protéique mais servent aussi de substrats dans d’autres mécanismes biologiques. Chez les bactéries, des aa-ARNt-transférases (AAT), possédant un domaine DUF2156, transfèrent l’acide aminé (aa) d’un aa-ARNt sur un glycérolipide. Ce remodelage membranaire participe à leur virulence et leur pathogénicité. Bien qu’un tel mécanisme n’ait jamais été démontré auparavant chez les eucaryotes, mon travail de thèse a révélé que de nombreux champignons possèdent des DUF2156/AAT qui aminoacylent des stérols. Ainsi, l’ergostéryl-aspartate et l’ergostéryl-glycine forment une nouvelle classe de lipides ; les stérols aminoacylés (SA). Mes travaux ont aussi permis de mettre en évidence une hydrolase catalysant la deacylation de certains SA. Aux vues du rôle central des stérols, nous proposons que ces voies de biosynthèse pourraient participer au remodelage membranaire, à la signalisation, à la résistance aux stresses ou à la pathogénie.</dcterms:abstract>
<dcterms:abstract xml:lang="en">Aminoacyl-tRNAs (aa-tRNA) play a central role in protein synthesis but can also be rerouted to other biological pathways. In bacteria, aa-tRNA transferases (AAT) having a DUF2156 domain catalyze the aminoacid (aa) transfer onto glycerolipids to improve their drug resistance or virulence. Such a mechanism was never described in eukaryotes. My thesis work revealed that numerous fungi species have DUF2156/AAT enzymes that aminoacylate sterols. Thus, ergosteryl-aspartate and ergosteryl-glycine are the founding members of a new class of lipids, namely aminoacylated sterols (AS). Moreover, we also identified a specific hydrolase that removes the aa from several AS. Considering the central role of sterols, we propose that AS might participate to cell surface remodeling, trafficking, resistance to stresses and/or pathogenicity.</dcterms:abstract>
