<dcterms:abstract xml:lang="fr">La leptine est une hormone anorexigène sécrétée par le tissu adipeux qui est acheminée vers l’hypothalamus pour induire la satiété. Elle est transportée du sang vers le liquide céphalo-rachidien en traversant les tanycytes de l’éminence médiane, des cellules épendymaires spécialisées qui bordent le 3e ventricule. Or, une des hypothèse mise en avant est que l’altération de ce transport participerait à la mise en place d’une résistance centrale à la leptine, un phénomène que l’on retrouve chez les patients atteints d’obésité. Il est donc primordial de mieux comprendre les mécanismes de transport des hormones métaboliques vers l’hypothalamus, un processus jusqu’alors inexploré au niveau cellulaire. Au cours de ma thèse, j’ai principalement étudié les mécanismes moléculaires qui régissent le transport tanycytaire de la leptine, de son internalisation jusqu’à sa sécrétion.</dcterms:abstract>
<dcterms:abstract xml:lang="en">Leptin is a hormone secreted by adipose tissue that acts in the central nervous system to regulate appetite. The dialogue between the periphery and the brain is essential to maintain energy homeostasis and involves tanycytes, specialized ependymal cells forming the wall of the 3nd ventricle. One of the hypotheses put forward is that alteration of this transport would contribute to the development of central leptin resistance, a phenomenon found in obese patients. It is therefore essential to better understand the mechanisms of transport of metabolic hormones to the hypothalamus, a process that has been unexplored until now at the cellular level. During my thesis, I mainly studied the molecular mechanisms governing the tanycytic transport of leptin, from its internalization to its secretion. Altogether, these data constitute a promising start toward the understanding of the leptin journey in tanycytes.</dcterms:abstract>
