<dcterms:abstract xml:lang="fr">Un mouvement coordonné repose non seulement sur une commande motrice appropriée, mais aussi sur l'analyse en direct des informations multi-sensorielles reçues. Le tractus cortico-spinal (CST) est soupçonné d’être responsable de ces fonctions. Le but de ma thèse a été de comprendre comment le contrôle cortical des entrées sensorielles s’organise autour de la commande motrice pour produire des comportements coordonnés. En étudiant les voies corticofuges, nous avons découvert que les contrôles moteur et sensoriel proviennent de la même zone corticale mais suivent ensuite des voies séparées. Nous avons montré que la commande motrice des membres postérieurs est principalement relayée par les centres moteurs supra-spinaux sous-corticaux, et n'est que partiellement encodée par le CST via un relais proprio-spinal rostral. En revanche, les neurones cortico-spinaux sont essentiels pour le contrôle sensoriel, qu’ils réalisent directement par l'intermédiaire d'une population d'interneurones lombaires. Cette nouvelle compréhension des fonctions de la voie cortico-spinale est fondamentale pour l’appréhension des maladies affectant le CST.</dcterms:abstract>
<dcterms:abstract xml:lang="en">A coordinated movement relies not only on a proper motor command, but also on the online analysis of a constant multi-sensory feedback. It has recently been hypothesized that the corticospinal tract (CST) could be responsible for such functions. The goal of my thesis was to unravel how the cortical filtering of sensory inputs hinges on the cortical motor command to produce well-coordinated behaviors. By sequentially investigating different stages of the corticofugal circuits, we found that both the motor and sensory control both originate from the same cortical area but then follow segregated paths. We showed that the hindlimb motor command is mainly relayed through subcortical supraspinal motor centers, and is only partly encoded by the CST via a propriospinal relay in the rostral spinal cord. In contrast, corticospinal neurons are essential for sensory control and execute it directly through a population of lumbar interneurons. This new understanding of the functions of the CST is fundamental for the study of diseases such as amyotrophic lateral sclerosis.</dcterms:abstract>
