<dcterms:abstract xml:lang="fr">La poly(ADP)-ribose polymérase PARP3 décrite dans des processus de progression tumorale a ici été étudiée dans la tumorigénèse d’un cancer agressif ayant un pronostic de survie très faible pour les patients, le glioblastome. Nous avons démontré une interaction de PARP3 avec le complexe de répression transcriptionnelle G9a/GLP1/Wiz et mis en évidence un axePARP3-G9a dans le contrôle de l’expression de gènes de la progression tumorale. D’autre part, nous avons observé une induction de PARP3 en conditions hypoxiques caractéristiques des glioblastomes. Par plusieurs expériences, nous avons démontré une faiblesse de la réponse cellulaire à l’hypoxie en absence de PARP3 et surtout, nous avons observé une fragilité exacerbée des microtubules dans des cellules PARP3-/- (CRISPR/Cas9). Nous avons alors exposé nos modèles cellulaires aux vinca-alcaloides. Les résultats obtenus ont validé une sensibilité des cellules PARP3-/- à ces molécules et une amplification de cette sensibilité en hypoxie. Ces résultats révèlent de nouvelles fonctions de PARP3 et font d’elle une cible prometteuse dans des stratégies anti-cancéreuses contre l’hypoxie dans le glioblastome.</dcterms:abstract>
<dcterms:abstract xml:lang="en">The poly(ADP)-ribose polymerase PARP3 described in tumour progression processes was here studied in the tumorigenesis of an aggressive cancer with a very poor survival prognosis for patients, glioblastoma. We demonstrated a physical and functional interaction of PARP3 with the G9a/GLP1/Wiz transcriptional repression complex and highlighted a PARP3-G9a axis in the control of several tumour progression genes expression. On the other hand, we observed an induction of PARP3 under hypoxic conditions characteristic of glioblastomas. In several experiments, we demonstrated a weak cellular response to hypoxia in the absence of PARP3 and, more importantly, we observed an exacerbated fragility of microtubules in PARP3-/- cells (CRISPR/Cas9). We then exposed our cell models to vinca alkaloids. The results obtained validated a sensitivity of PARP3-/- cells to these molecules and an amplification of this sensitivity under hypoxia. These results reveal new functions of PARP3 and make it a promising target for anti-cancer strategies against hypoxia in glioblastoma.</dcterms:abstract>
