<dcterms:abstract xml:lang="fr">Les C-glycosides représentent l’une des classes de glycomimétiques la plus importante et la plus étudiée. Le remplacement de l’atome d’oxygène exocyclique anomérique d’O-glycosides par un atome de carbone a pour conséquence une augmentation de la résistance à l’hydrolyse enzymatique in vivo. Au sein de cette famille, les bis-C-glycosides sont caractérisés par la présence de deux substituants carbonés en position pseudo anomérique formant un centre quaternaire.L’objectif de ce travail de thèse a été de mettre au point une nouvelle approche catalytique de la synthèse de bis-C-glycosides, à partir d’exo-glycals, via la génération de radicaux pseudo anomériques tertiaires. Ces radicaux, généré par transfert d’atome d’hydrogène depuis un hydrure de fer (III), ont alors été additionnés sur des alcènes électroappauvris pour conduire aux cibles visées. Une étude préliminaire a ensuite visé la préparation de dérivés bicycliques spiraniques et fusionnés par cyclisation sur des alcynes silylés. Les limitations rencontrées lors de cette étude nous ont conduits à mener une étude sur des énynes modèles, au cours de laquelle un processus d’abstraction intramoléculaire remarquablement efficace a été identifié. L’observation inattendue de ce dernier nous a finalement menés à conduire une première série d’expériences préliminaires au développement de nouvelles cascades radicalaires originales.</dcterms:abstract>
<dcterms:abstract xml:lang="en">C-glycosides are one of the most important and studied classes of glycomimetics. The substitution of the exocyclic anomeric oxygen atom of O-glycosides by a carbon atom results in an increase of in vivo resistance to enzymatic hydrolysis. Within this family, bis-C-glycosides are characterized by the presence of two carbon substituents on the pseudo-anomeric position, establishing a quaternary centre.The aim of this PhD work was to develop a new catalytic approach to the synthesis of bis-C-glycosides, via the intermediacy of tertiary pseudo-anomeric radicals, from exo-glycals. These radicals, generated by hydrogen atom transfer from iron (III) hydride, could be added to electron-poor alkenes to lead to the target molecules. A preliminary study then led us to the preparation of spiro and fused bicyclic glycomimetics by radical cyclisations on silylated alkynes. Some limitations guided us to conduct studies on enynes models, during which a remarkably efficient intramolecular abstraction process was identified. This study sets the stage for the development of original, sustainable radical cascade reactions.</dcterms:abstract>
