<dcterms:abstract xml:lang="fr">La voie pro-oxydante de l’angiotensine II est associée à une surexpression des co-transporteurs sodium glucose 1 et 2 (SGLT1 et 2) au sein des cellules endothéliales ainsi qu’à l’induction de la dysfonction endothéliale dans les artères coronaires. L’ensemble de ces effets a été prévenu par l’inhibiteur mixte de SGLT1 et 2, sotagliflozine ainsi que par l’inhibiteur sélectif de SGLT2, l’empagliflozine suggérant ainsi l’implication de ces transporteurs dans l’induction de la dysfonction endothéliale. L’objet de cette étude est d’évaluer le rôle de SGLT1 et 2 dans la dysfonction vasculaire et cardiaque induites par la réponse hypertensive de l’angiotensine II chez le rat. L’étude montre que l’hypertension induite par l’angiotensine II mène à une dysfonction diastolique du ventricule gauche et à une dysfonction endothéliale des macro et des micro vaisseaux associées à une surexpression de SGLT1 et 2 dans les cellules endothéliales. De plus, ce travail indique que l’ensemble des effets cardiovasculaires délétères observés sont prévenus par l’empagliflozine sans pour autant moduler la pression artérielle. L’ensemble de cette étude montre que l’inhibition des transporteurs SGLT1/2 apparait comme une intéressante stratégie pour protéger la fonction CV des patients non diabétiques avec dysfonction diastolique.</dcterms:abstract>
<dcterms:abstract xml:lang="en">The angiotensin II prooxidant pathway has been shown to upregulate the expression of SGLT1 and 2 and to induce premature endothelial dysfunction in coronary endothelial cells. Since these effects are prevented by the dual SGLT1/2 inhibitor sotagliflozin and the SGLT2 inhibitor empagliflozin, SGLT1 and 2 are involved in the induction of endothelial dysfunction. The aim of the study is to evaluate the rôle of SGLT1 and 2 in the Ang II-induced hypertensive response in rats leading to cardiac and vascular endothelial dysfunction. The findings indicate that Ang II-induced hypertension promoting left ventricle cardiac dysfunction, and both macro and microvascular endothelial dysfunction are associated with an up-regulation of SGLT1 and 2 in endothelial cells. They further suggest that inhibition of SGLT1 and/or SGLT2 might be an attractive therapeutic strategy to protect the CV function, and, hence, the subsequent development of cardiovascular diseases.</dcterms:abstract>
