<dcterms:abstract xml:lang="fr">De nombreux aspects interviennent dans la régulation de la thrombopoïèse. La composante mécanique de l’environnement médullaire fait partie des facteurs à considérer lors de l’étude de la formation des plaquettes sanguines. Ce travail de thèse avait pour objectif d’identifier les mécanismes par lesquels les mégacaryocytes s'adaptent aux changements de contraintes mécaniques de leur environnement. Le projet portait sur l’impact de la perte de confinement qui survient lors de l’extension des proplaquettes dans la circulation sanguine. Les travaux réalisés ont mis en avant une différence d’implication du cytosquelette dans l’extension des proplaquettes entre in vitro et in vivo. D’autre part, mes travaux ont mis en évidence un rôle majeur du calcium extracellulaire dans la formation des proplaquettes. Les résultats obtenus permettent également de conclure d’une implication, au moins partiellement mécano-dépendante, du canal TRPV4 dans ce phénomène. De plus, un rôle de TRPV2 et des récepteurs Piezo 1 et 2 dans la thrombopoïèse a été démontré.</dcterms:abstract>
<dcterms:abstract xml:lang="en">Many aspects are involved in the regulation of thrombopoiesis. The mechanical properties of the medullar environment are to be considered when studying the production of blood platelets. The aim of this thesis was to identify the mechanisms by which megakaryocytes adapt to mechanical changes in their environment. The project focused on the impact of the loss of confinement that occurs during the proplatelet extension in the bloodstream. The work carried out highlighted a difference in the involvement of the cytoskeleton in proplatelet formation between in vitro and in vivo conditions. On the other hand, my work has highlighted a major role of extracellular calcium in proplatelet formation. The results obtained allow us to conclude that the TRPV4 channel is involved mechanically, at least in part, in the process. It also demonstrates a role for TRPV2 and Piezo 1 and 2 receptors in thrombopoiesis.</dcterms:abstract>
