<dcterms:abstract xml:lang="fr">Un défi majeur posé aux vecteurs d’acides nucléiques dans le poumon est de traverser le mucus, afin d’accéder aux cellules devant être transfectées. Dans cette problématique, nous avons cherché à développer des vecteurs mucolytiques, i.e. capables de déstructurer localement les ponts disulfures des mucines, pour une meilleure diffusion à travers le mucus, et donc une meilleure efficacité de transfection. Des modèles cellulaires producteurs de mucus ont été mis en place pour une évaluation pertinente de ces vecteurs. Les premiers vecteurs mucolytiques évalués étaient des lipides cationiques monosoufrés, pour lesquels ni activité mucolytique ni bénéfice en termes de transfection n’ont pu être mis en évidence. Dans une deuxième stratégie, des lipides cationiques ont été associés à des lipides helper polysoufrés à plus haut potentiel mucolytique. Ces nouvelles formulations possédaient une activité mucolytique, diffusaient plus rapidement et plus profondément dans le mucus, et avaient une efficacité de transfection augmentée. Ce travail est le premier à introduire le concept de vecteurs d’acide nucléique à activité mucolytique intrinsèque capables de déstructurer localement le mucus, et suggère que cette stratégie est encourageante, en particulier au regard de l’utilisation d’agents mucolytiques tels que la N-acétylcystéine ayant une action généralisée sur le mucus.</dcterms:abstract>
<dcterms:abstract xml:lang="en">A major challenge for nucleic acid carriers in the lung is to cross the mucus to reach the epithelial cells in order to transfect them. In this issue, we sought to develop mucolytic carriers, i.e. able to open the mesh structure of the mucus by breaking the disulfide bridges of the mucins, to improve their diffusion through the mucus, and thus increase their transfection efficiency. Mucus-producing cellular models were set up for a relevant evaluation of those carriers. The first mucolytic carriers to be evaluated were monothiolated cationic lipids, for which no mucolytic activity nor gain in transfection activity could be highlighted. In a second strategy, cationic lipids were associated to polythiolated helper lipids with higher mucolytic potential. Those new formulations held a mucolytic activity, diffused faster and deeper in the mucus, and had an increased transfection efficiency. This work is the first to introduce the concept of gene carrier with an intrinsic mucolytic activity able to break down the mucus locally, and suggest that this strategy is promising, in particular compared to the use of mucolytic agents such as N-acetylcysteine having a generalized action on the mucus.</dcterms:abstract>
