<dcterms:abstract xml:lang="fr">La réponse immunitaire innée, la première ligne de défense contre les infections, est conservée à travers les métazoaires. En revanche, une dérégulation de celle-ci entraîne une inflammation chronique délétère. Mon travail de doctorat consistait en l’étude de la régulation des voies NF-kB, une voie majeure de l’inflammation. En utilisant le modèle Drosophila melanogaster, j’ai étudié la régulation des voies NF-kB à différents niveaux. Un objectif majeur de ma thèse a été d’explorer le rôle des longs ARN non-codants dans la régulation des voies NF-kB, alors que d’autres aspects de ma thèse ont été d’explorer leur régulation par des ubiquitine-ligases, des micro ARNs, des phosphatases ou des modulateurs épigénétiques. J’ai utilisé différents outils expérimentaux tels que le système CRISPR Cas9 pour évaluer la fonction de gènes candidats. Nous pensons que ce travail pourrait ouvrir la voie à de nouvelles cibles thérapeutiques pour le traitement de maladies liées au système immunitaire.</dcterms:abstract>
<dcterms:abstract xml:lang="en">The innate immune system is well conserved across metazoan species, enabling them to fight off invading microbes. However, aberrant activation of innate immune responses causes deleterious chronic inflammation. The core of my PhD work was to study the fine-tuned regulation of the NF-kB pathway, one of the major pathways controlling inflammation. Using the model organism Drosophila melanogaster, I studied the regulation of NF-kB pathways at various levels. One focus of my PhD has been to uncover the role of long non-coding RNAs in the regulation of NF-kB pathways, while other aspects of my thesis revolved around the exploration of NF-kB pathway regulation by E3 Ubiquitin ligases, micro RNAs, phosphatases, or epigenetic regulators. I used several experimental tools including genetic tools such as the CRISPR Cas9 system to explore the function of candidate genes. We believe that this work could lead to potential therapeutic targets for the treatment of immune-related diseases.</dcterms:abstract>
