<dcterms:abstract xml:lang="fr">Les informations nociceptives en provenance de la périphérie sont traitées dans la corne dorsale de la moelle épinière, puis elles sont projetées aux centres supra-spinaux où la perception douloureuse pourra être élaborée. L’objectif de ce travail de thèse était d’étudier les phénomènes de plasticité synaptique à court-terme (STP) des synapses inhibitrices entre les neurones de la lamina II de la corne dorsale de la moelle épinière en réalisant des enregistrements électrophysiologiques. Les résultats obtenus indiquent 1) que les connexions GABAergiques entre les neurones de la lamina II expriment des mécanismes de STP qui diffèrent suivant la fréquence de stimulation du neurone présynaptique, 2) ainsi que selon la nature du neurone postsynaptique, 3) que ces plasticités mettent en jeu l’activation de récepteurs GABAB et/ou A1 et 4) que ces plasticités sont modifiées en conditions d’inflammation périphérique. Les STP mises en évidence pourraient avoir un impact sur la balance excitation/inhibition. Ces plasticités et leur modulation par les récepteurs GABAB et A1 pourraient représenter des cibles pour le développement de stratégies analgésiques.</dcterms:abstract>
<dcterms:abstract xml:lang="en">Networks of the dorsal horn of the spinal cord process nociceptive information from the periphery. Using electrohysiological recordings, the present work aimed to define the short-term plasticity (STP) of lamina II neurons inhibitory connections. The results obtained indicate 1) that GABAergic connections between the lamina II neurons express STP mechanisms that differ according to the frequency of stimulation of the presynaptic neuron, 2) as well as according to the nature of the postsynaptic neuron, 3) that these plasticities involve the activation of GABAB and/or A1 receptors and 4) that those plasticities are modified under peripheral inflammatory conditions.Target-specific and frequency-specific plasticities could then tune the balance between inhibition and disinhibition while processing frequency-coded information from primary afferents. These short-term plasticities and their modulation by A1 and GABAB receptors might represent an interesting target in pain-alleviating strategies.</dcterms:abstract>
