<dcterms:abstract xml:lang="fr">Jusqu’à maintenant, la synthèse peptidique en phase solide (SPPS) était utilisée en suivant le concept développé par Merrifield en 1963 : avec un large excès de réactifs et de solvants, en contradiction avec les 12 principes de la chimie verte. Durant cette thèse, l’étape de préactivation a été optimisée dans le DMF en réduisant les excès de réactifs, et des temps de préactivation plus longs ont mené à des couplages performants en un temps réduit. Ensuite, des solvants et mélanges de solvants alternatifs ont été testés pour remplacer le DMF, et les études cinétiques de préactivation, couplage et déprotection nous ont permis de sélectionner certains mélanges de solvants. La déprotection du Fmoc dans ces mélanges fût ensuite optimisée avec des bases alternatives, et le nombre de lavages après déprotection fût réduit. Ces conditions optimisées ont été utilisées pour la synthèse de différents peptides modèles avec différentes résines et charges. Les temps de couplage, puretés et rendements obtenus avec les faibles charges étaient comparables à ceux dans le DMF, mais des optimisations seront nécessaires avant de pouvoir envisager l’implémentation en production.</dcterms:abstract>
<dcterms:abstract xml:lang="en">Up-to-now, the solid phase peptide synthesis (SPPS) has been used following the concept designed by Merrifield in 1963: large excess of reagents and solvents are used, which is in contradiction with the 12 Principles of Green Chemistry. During this PhD, the preactivation step was optimized in DMF with lower excess of reagents, and longer preactivation times led to efficient couplings in reduced time. Then alternative solvents and solvent mixtures were tested to replace DMF, and kinetic studies of preactivation, coupling reaction, and deprotection led us to select some mixtures of alternative solvents. Fmoc deprotection in these specific media was then optimized with alternative bases, and the number of washes after the deprotection was reduced. The resulting optimized conditions were employed for the synthesis of different model peptides with different resins and loadings. Coupling times, purities and yields obtained with low loadings in alternative solvents were competitive with DMF, but some optimizations are still required before considering industrial-scale implementation.</dcterms:abstract>
