<dcterms:abstract xml:lang="fr">Ces travaux de thèse ont permis de développer des biomarqueurs fluorescents (BMFs) dans le but de créer un dispositif de diagnostic des pathologies cornéennes. Le BMF est constitué d’un fluorophore lié par un linker à un anticorps. Le fluorophore est un BODIPY et l’anticorps un nanobody (ou VHH). Une première partie de ces travaux a permis d’obtenir une bibliothèque d’architectures fluorophores-linker et de mettre au point la conception et l’élaboration du biomarqueur allant de la synthèse du fluorophore jusqu’à la bioconjugaison avec le nanobody. Pour cela un fluorophore modèle, le BOD F, a été utilisé ainsi qu’un anticorps modèle le VHH72c. Les BMFs obtenus sont toujours capables de reconnaitre leur antigène et la fluorescence du BODIPY n’est pas éteinte en milieu physiologique (tampon PB pH 7,4). Une deuxième partie de ces travaux a consisté à développer des fluorophores et donc des BMFs qui absorbent et émettent dans le rouge et le proche infrarouge. Pour celà le BOD T émettant à 650 nm a été fonctionnalisé et bioconjugué au VHH72c. Un nouveau fluorophore le BOD TT émettant à 700 nm a également été synthétisé, fonctionnalisé et bioconjugué au VHH72c. En parallèle des travaux ont également été réalisés dans le but de développer des fluorophores hydrosolubles afin de faciliter l’étape de bioconjugaison.</dcterms:abstract>
<dcterms:abstract xml:lang="en">This thesis project allowed the development of fluorescent biomarkers (BMFs) aiming to create a diagnostic device for corneal pathologies. The BMF consists of a fluorophore bound by a linker to an antibody. The fluorophore is a BODIPY and the antibody is a nanobody (or VHH). The first part of this work made it possible to obtain a library of fluorophore-linker architectures and to develop the design and elaboration of the biomarker, from the synthesis of the fluorophore to the bioconjugation with the nanobody. For this purpose, a model fluorophore BOD F was used as well as a model antibody VHH72c. The BMFs obtained are still capable of recognising their antigen and the BODIPY fluorescence is not extinguished in physiological medium (PB buffer pH 7.4). A second part of this work consisted in developing fluorophores and thus BMFs that absorb and emit in the red and near infrared regions. To this end, BOD T emitting at 650 nm was functionalized and bioconjugated with VHH72c. A new fluorophore, BOD TT, emitting at 700 nm was also synthesized, functionalised and bioconjugated with VHH72c. In parallel, a part of this work was dedicated to the development of water-soluble fluorophores to facilitate the bioconjugation stage.</dcterms:abstract>
