L’analyse protéomique par spectrométrie de masse permet l’étude de systèmes biologiques complexes en identifiant et quantifiant les protéines impliquées et en caractérisant leur structure. Cette stratégie peut aussi être appliquée à l’étude des modifications post-traductionnelles des protéines, niveau de complexité supplémentaire du protéome. Ce travail de thèse est axé sur la mise en place de stratégies analytiques quantitatives globales de type « label-free » pour explorer les mécanismes biologiques à l’œuvre chez l’ours brun et le lérot hibernants. D’autre part, des développements pour l’analyse quantitative des phosphorylations dans des tissus musculaires et des glycations dans des échantillons plasmatiques ont été réalisés, de la préparation des échantillons avec notamment des stratégies d’enrichissement par chromatographies d’affinité et de fractionnement peptidique, jusqu’au traitement des données de spectrométrie de masse par diverses solutions logicielles.
Proteomic analysis by mass spectrometry enables the study of complex biological systems by identifying and quantifying their proteins and characterising their structure. This strategy can also be applied to the study of post-translational modifications, an additional level of complexity in the proteome. This thesis work focuses on the implementation of global quantitative label-free strategies to explore the biological mechanisms at work in the hibernating brown bear and garden dormouse. On the other hand, developments for the quantitative analysis of phosphorylations in muscle tissues and glycations in plasma samples were carried out, from the sample preparation with enrichment strategies by affinity chromatography and peptide fractionation, to the processing of mass spectrometry data by several software solutions.
