<dcterms:abstract xml:lang="fr">Lors de cette thèse, nous avons conçu et développé de nouvelles approches basées sur des réactions d’hétérocycloaddition pour la diversification structurale et le marquage de molécules à visées thérapeutique et diagnostique. Nous avons d’abord appliqué la réaction de Povarov à la diversification du squelette aza-dicétopipérazine, une famille de N-hétérocycles sous-représentés dans la littérature. Une vingtaine de structures tétracycliques originales ont été obtenues sous la forme de paires séparables de diastéréoisomères. Par la suite, des aza-dicétopipérazines analogues de dicétopipérazines bioactives ont également été synthétisées dans le but d’identifier des activités biologiques au sein de cette nouvelle famille d’hétérocycles. Dans une seconde partie, nous nous sommes intéressés à l’utilisation d’une réaction de thia-Diels-Alder impliquant un phosphonodithioester et un diène exocyclique hautement réactif pour le marquage chimiosélectif de peptides. Cette cycloaddition s'est montrée rapide et efficace nous permettant de l'utiliser avec succès sur différents peptides pour des applications en imagerie nucléaire (Tomographie par Emission de Positons, 18F) et en imagerie optique (fluorophores). Enfin, différentes stratégies « double click » impliquant cette réaction de thia-Diels-Alder ont été étudiées et ont montré des résultats encourageants.</dcterms:abstract>
<dcterms:abstract xml:lang="en">During this thesis, we designed and developed new strategies based on heterocycloaddition reactions for the structural diversification and the labelling of molecules with therapeutic and diagnostic applications. We first applied the Povarov reaction to the diversification of the aza-diketopiperazine scaffold, a family of N-heterocycles scarcely represented in the literature. Around twenty original tetracyclic structures were obtained as pairs of separable diastereomers. We then aimed to discover biological activities amongst these N-heterocycles by synthesising aza-diketopiperazine-analogues of bioactive diketopiperazines. In a second part, we focused on the application of a thia-Diels-Alder reaction involving a phosphonodithioester and a highly reactive exocyclic diene to the chemoselective labelling of peptides. This reaction was fast and efficient which led to its successful use on several peptides with fluorine-18 for in vivo Positron Emission Tomography (PET) or fluorophores for in cellulo optical imaging. Finally, a couple a “double click” strategies involving our thia-Diels-Alder reaction were studied and showed promising préliminary results.</dcterms:abstract>
