<dcterms:abstract xml:lang="fr">La dernière décennie marque d'importantes évolutions dans le domaine de la biologie structurale, en particulier dans la cristallographie sérielle aux rayons X résolue en temps. L’objectif de cette thèse en cotutelle entre les universités de Strasbourg et de Leipzig, était d’appliquer cette approche à la caractérisation fonctionnelle et structurale de trois CCA-adding enzymes. En combinant résultats biochimiques et méthodes de cristallisation avancées, nous avons pu : 1) déterminer l'affinité de liaison de nos candidats à divers substrats ainsi que les propriétés cinétiques des réactions catalysées , 2) obtenir des conditions reproductibles de production de cristaux pour les analyses standard et SSX,3) suivre les événements de nucléation et contrôler la cristallisation de certaines enzymes grâce au Xtal Controller, 4) acquérir des données sérielles in situ à température ambiante en puce microfluidique, 5) résoudre la structure d’une cible dans sa forme apo ou en complexe avec un ligand.</dcterms:abstract>
<dcterms:abstract xml:lang="en">The last decade has seen important developments in the field of structural biology, in particular in time-resolved serial X-ray crystallography (TR-SSX). The objective of this thesis, in cotutelle between the universities of Strasbourg and Leipzig, was to apply this approach to the functional and structural characterization of three CCA-adding enzymes. By combining biochemical results and advanced crystallization methods, we were able to: 1) determine the binding affinity of our candidates to various substrates as well as the kinetic properties of the catalyzed reactions, 2) obtain reproducible crystal production conditions for standard and SSX analyses, 3) monitor nucleation events and control the crystallization of selected enzymes using the Xtal Controller, 4) acquire serial data in situ at room temperature in microfluidic chip, 5) resolve the structure of a target in its apoform or in complex with a ligand.</dcterms:abstract>
