<dcterms:abstract xml:lang="fr">Afin d’achever leur cycle de réplication, les virus qui accèdent au noyau de la cellule hôte, doivent exporter des ARN épissés ou non vers le cytoplasme. L'étude des rétrovirus de mammifères a conduit à la découverte de la voie d'export principale des ARNm cellulaire, basée sur les récepteurs d'export TAP‑p15 et le complexe TREX-1, qui, à son tour est recruté lors des étapes de maturation de l’ARNm. Certains virus, tel que le VIH-1, usurpent aussi une voie d’export alternative en recrutant XPO1, la principale exportine des protéines, sur leurs ARN. Chez les plantes, les mécanismes précis d'export nucléaire des ARNm cellulaires sont moins bien connus, et, jusqu'à récemment, aucune donnée n'était disponible sur l'export nucléaire de l'ARN d'un phytovirus. Avec des résultats précédents, les travaux présentés dans ce manuscrit montrent que, à l'instar du VIH‑1, le (pararétro)virus de la mosaïque du chou-fleur, peut exploiter les deux voies d’export, TREX-1 et XPO1, pour exporter différentes isoformes de son ARN 35S du noyau. Ce mécanisme implique à la fois la protéine de capside P4 et la transcriptase inverse P5, ainsi que des facteurs d’export cellulaires.</dcterms:abstract>
<dcterms:abstract xml:lang="en">To complete their replication cycle, viruses accessing the nucleus of the host cell must export spliced and unspliced RNAs to the cytoplasm. Studying mammalian retroviruses led to the discovery of the major cellular mRNA export pathway, based on the TAP-p15 export receptors and the TREX-1 complex, which is recruited during the mRNA maturation steps. Some viruses, such as HIV-1, also hijack an alternative export pathway by recruiting XPO1, the major protein exportin, to their RNA. In plants, the precise export mechanisms for cellular mRNA are less well known, and until recently, no data was available on the nuclear export of any plant virus RNA. Together with previous results, the work presented in this manuscript shows that, not unlike HIV-1, the cauliflower mosaic (pararetro)virus exploits both the TREX-1 and XPO1 export pathways to export different isoforms of its 35S RNA from the nucleus. The mechanism involves both the viral capsid protein P4 and the reverse transcriptase P5, as well as cellular export factors.</dcterms:abstract>
