<dcterms:abstract xml:lang="fr">La production de plaquettes de culture est aujourd’hui possible mais est limitée à la génération d’une centaine de plaquettes par mégacaryocyte (MK) quand, dans la moelle, un MK serait capable de produire 2000 à 3000 plaquettes. Si les connaissances ont fortement progressé sur les mécanismes moléculaires et cellulaires qui gouvernent la mégacaryopoïèse, les mécanismes qui régissent la libération de plaquettes sont encore peu connus. Dans ce contexte, le laboratoire que j’ai intégré a développé deux dispositifs de libération des plaquettes qu’il était nécessaire de caractériser et d'optimiser. Ce travail de thèse avait pour objectif de de caractériser ces deux dispositifs tout en analysant l’effet des écoulements produits sur la libération plaquettaire. De par ces travaux, nous avons confirmé la nécessité de la présence de flux turbulents dans la libération de plaquettes et nous avons également mis en lumière un nouveau paramètre qu’il serait intéressant de prendre en compte dans le développement de futurs dispositifs : le couplage entre l’intensité et la durée de la contrainte de cisaillement, un tandem favorisant la rupture membranaire.</dcterms:abstract>
<dcterms:abstract xml:lang="en">The production of cultured platelets is today possible, but it is limited to the generation of about 100 platelets per megakaryocyte when, in the marrow, a megakaryocyte would be capable of producing 2000 to 3000 platelets. Although knowledge has progressed considerably on the molecular and cellular mechanisms governing megakaryopoiesis, the mechanisms governing platelet release are still poorly understood and, as a result, the platelet release devices developed are still not very effective. In this context, the laboratory I joined had developed two platelet release devices that needed to be characterized to allow their optimization. The objective of this thesis was to characterize these two devices while analyzing the effect of the flows produced on the platelet release. Through this work, we confirmed the necessity of turbulent flows in platelet release and we also highlighted a new parameter that would be interesting to take into account in the development of future devices; the coupling between the intensity and duration of the shear stress, a tandem that favors platelet rupture.</dcterms:abstract>
