<dcterms:abstract xml:lang="fr">La plupart des infections causées par Candida albicans - l'agent pathogène fongique le plus courant - sont associées à des traitements très toxiques et coûteux. Le principal défi aujourd'hui est de trouver les inhibiteurs spécifiques qui agissent uniquement contre la cible choisie, comme le ribosome de C. albicans. C. albicans a été observé empiriquement dans les hôpitaux comme étant résistant au cycloheximide, qui se lie au E-site de liaison de l'ARNt sur la sous-unité 60S. Pour la première fois, nous avons obtenu la structure à haute résolution du ribosome de C. albicans (2,3 Å) par microscopie électronique cryogénique et proposé le mécanisme de résistance au cycloheximide chez C. albicans. Nos résultats offrent une base solide pour le développement futur potentiel de médicaments qui se lient spécifiquement au ribosome de C. albicans.</dcterms:abstract>
<dcterms:abstract xml:lang="en">Most infections caused by Candida albicans - the most common fungal pathogen - are associated with very toxic and expensive treatments. The main challenge nowadays is to find the specific inhibitors that only act against the chosen target, such as the C. albicans ribosome. C. albicans has been empirically observed in hospitals to be resistant to cycloheximide, which binds to the E-tRNA binding site on the 60S subunit. For the first time, we obtained the high-resolution structure of C. albicans ribosome (2.3 Å) by cryogenic electron microscopy and proposed the mechanism of cycloheximide resistance in C. albicans. Our results offer a solid basis for the potential future development of drugs that specifically bind to the C. albicans ribosome.</dcterms:abstract>
