<dcterms:abstract xml:lang="fr">Le trafic intracellulaire du cholestérol est essentiel à l’homéostasie du cholestérol et cellulaire. Les mécanismes de régulation de l’export du cholestérol hors du lysosome sont encore peu détaillés. Nous avons mis en évidence un nouveau rôle de la signalisation Wnt dans l’homéostasie du cholestérol.En effet, l’absence du ligand Wnt5a entraine une activation de la signalisation mTORC1, une accumulation de cholestérol dans le lysosome, une dysfonction lysosomale et favorise le développement de l’athérosclérose. Wnt5a augmente l’expression de la lipase acide lysosomale (LAL), interagit avec Niemann-Pick C1 (NPC1) et Niemann-Pick C2 (NPC2), deux protéines lysosomales transportant le cholestérol lysosomal, et diminue l’activité du complexe mTORC1. Wnt5a communique ainsi le taux de cholestérol en diminuant l’activité de mTORC1, favorisant la sortie du cholestérol hors du lysosome vers le RE, limitant l’accumulation de cholestérol et protégeant contre l’athérosclérose.Ainsi, Wnt5a est un nouvel élément essentiel à la régulation du trafic intracellulaire du cholestérol.</dcterms:abstract>
<dcterms:abstract xml:lang="en">Intracellular cholesterol trafficking is essential for cholesterol and cellular homoeostasis. The mechanisms involved in the regulation of the export of cholesterol from the lysosome are still poorly understood. We have identified a novel role for Wnt signalling in cholesterol homoeostasis.Indeed, the absence of Wnt5a ligand leads to the activation of mTORC1 signalling, lysosomal cholesterol accumulation, lysosomal dysfunction and promotes the development of atherosclerosis. Wnt5a increases the expression of lysosomal acid lipase (LAL), interacts with Niemann-Pick C1 (NPC1) and Niemann-Pick C2 (NPC2), two lysosomal cholesterol-transporting proteins, and decreases mTORC1 signalling. Thus, Wnt5a communicates cholesterol levels by decreasing the activity of mTORC1, promoting cholesterol egress from the lysosome to the ER, limiting cholesterol accumulation and protecting against atherosclerosis.Thus, Wnt5a is an essential component of the lysosomal cholesterol export machinery</dcterms:abstract>
