<dcterms:abstract xml:lang="fr">La Cohésine, complexe à 4 sous-unités (SMC1A, SMC3, RAD21, STAG1/2), est un effecteur majeur de l’organisation 3D du génome eucaryote. Il participe à de nombreuses transactions avec l'ADN (cohésion des chromatides sœurs, réparation de l'ADN, régulation de la transcription et organisation 3D du génome). La Cohésine possède une activité ATPase essentielle à sa dynamique et à ses fonctions et qui est portée par SMC1A et SMC3. Ces protéines forment un hétérodimère en présence d’ATP (module ATPase). Le rôle de l’activité ATPase de la Cohésine sur sa dynamique et ses mécanismes moléculaires est peu compris. Au cours de ma thèse j’ai caractérisé les interactions entre SMC1A, SMC3 et RAD21 par des méthodes biophysiques, biochimiques et structurales. J’ai obtenu une structure cryo-EM à 4 Å du module ATPase. Je me suis également intéressée à la 4e sous-unité de la Cohésine STAG1/2 et son influence sur le module ATPase. Mes travaux de thèse révèlent des informations novatrices sur les mécanismes impliqués dans l’activité ATPase de la Cohésine, notamment au niveau de la porte de sortie de l’ADN, permettant ainsi une meilleure compréhension de sa dynamique et de ses fonctions.</dcterms:abstract>
<dcterms:abstract xml:lang="en">Cohesin, a four-subunit complex (SMC1A, SMC3, RAD21, STAG1/2), is a key effector of the 3D organization of the eukaryotic genome. It is involved in many transactions with DNA (sister chromatid cohesion, DNA repair, regulation of transcription and 3D genome organization). Cohesin has an ATPase activity which is essential to its dynamics and to all its functions and is carried by SMC1A and SMC3. These proteins form a heterodimer in the presence of ATP (ATPase module). The role of the ATPase activity of Cohesin on its dynamics and molecular mechanisms is poorly understood. During my thesis, I characterized the interactions between SMC1A, SMC3, and RAD21 by biophysical, biochemical, and structural methods. I obtained a 4 Å cryo-EM structure of the ATPase module. I also focused on the 4th subunit of the STAG1/2 cohesin and its influence on the ATPase module. My thesis work reveals novel information on the mechanisms involved in the ATPase activity of Cohesin, especially at the DNA exit gate, allowing a better understanding of its dynamics and functions.</dcterms:abstract>
