<dcterms:abstract xml:lang="fr">La méthylation de l’ADN est une marque épigénétique mise en place par les enzymes appelées méthyltransférases de l’ADN (DNMTs). DNMT1, DNMT3A et DNMT3B sont les trois principales DNMTs : DNMT3A et DNMT3B catalysent la méthylation de novo de l'ADN tandis que DNMT1 maintient les profils de méthylation au cours des divisions cellulaires. La méthylation de l’ADN est un phénomène dynamique qui connait deux reprogrammations majeures : l’une dans l’embryogenèse précoce au cours du développement et l’autre dans les cellules germinales primordiales (PGCs). Cela consiste en un effacement quasi-total de la méthylation puis reméthylation globale du génome. Dans ce travail de thèse, nous nous sommes intéressés à la contribution de DNMT3B dans la phase de méthylation des cellules germinales mâles et de DNMT3A dans la phase de reméthylation dans l’embryon en utilisant des lignées génétiquement modifiées de souris. Dans le projet 1, des difficultés expérimentales ne nous ont malheureusement pas permis de conclure quant à la contribution de DNMT3B dans la reméthylation des cellules germinales mâles. Dans le projet 2, j'ai confirmé le rôle de DNMT3A en générant des souris KO pour le gène. Nous avons mis en évidence une létalité embryonnaire partielle, des défauts de croissance après la naissance et des défauts de méthylation de l'ADN dans plusieurs organes. Nous avons également confirmé l’existence de deux isoformes de DNMT3A et mis en évidence le remplacement de l'isoforme DNMT3A2 par DNMT3A1 au cours du développement. En générant des souris KO pour Dnmt3a1, j'ai analysé le rôle spécifique de cette isoforme dans le développement et la méthylation de l'ADN. Enfin, nous avons étudié les modifications du transcriptome et de l’accessibilité de la chromatine induites par l'absence de DNMT3A. Ce travail met en lumière l'importance de la méthylation de l'ADN déposée par DNMT3A et la contribution des isoformes de DNMT3A pour la mise en place du transcriptome et de l'identité cellulaire au cours du développement chez la souris.</dcterms:abstract>
<dcterms:abstract xml:lang="en">DNA methylation is an epigenetic mark catalyzed by enzymes called DNA methyltransferases (DNMTs). DNMT1, DNMT3A and DNMT3B are the three main DNMTs and have different roles: DNMT3A and DNMT3B catalyze de novo methylation while DNMT1 is required for maintenance of DNA methylation through cell divisions. DNA methylation is a dynamic mark that undergoes two major waves of reprogramming: one in the embryo during early development and the other one in primordial germ cells (PGCs). They consist in an almost complete erasure of DNA methylation followed by global remethylation of the genome. In this thesis work, we investigated the role of DNMT3B in the global remethylation of the genome during male PGC development and the role of DNMT3A in the global remethylation of the genome in the embryo using genetically modified mice. In the first project, experimental difficulties did not allow us to conclude on the role of DNMT3B in male PGCs. In the second project, we highlighted the role of DNMT3A by generating Dnmt3a knock-out mouse models. We showed that the inactivation of Dnmt3a leads to partial embryonic lethality, growth defects leading to death and DNA methylation defects in various organs. We also confirmed the existence of two isoforms, DNMT3A2 and DNMT3A1, that are expressed successively during development. By generating novel mouse models carrying a specific knockout of Dnmt3a1, we analyzed the specific role of this isoform in development and DNA methylation. Finally, we studied the changes in transcriptome or chromatin accessibility caused by the absence of DNMT3A. This work highlights the importance of DNA methylation deposited by DNMT3A and the contribution of DNMT3A isoforms for the establishment of the transcriptome and cell identity during development in mice.</dcterms:abstract>
