<dcterms:abstract xml:lang="fr">Les e-cigarettes (e-cig) n’ont pas démontré leur efficacité et leur tolérance, en partie à cause d’un large choix de liquides (eL) contenant des véhicules, des arômes et de la nicotine à différentes concentrations, complexifiant les études scientifiques. Les travaux présentés ici concernent 3 niveaux d’analyse. Chez l’Homme, les résultats de l’essai clinique VAPexp’AIR sont les premiers à montrer que la formulation du eL influence la masse de nicotine inhalée et ses paramètres pharmacocinétiques. Chez la souris, un modèle de BPCO post-tabagique a été développé présentant des caractéristiques pathologiques telles que le recrutement des cellules immunitaires dans les voies aériennes inférieures associé à de l’emphysème pulmonaire. Ce modèle sera utilisé comme témoin lors de l’exposition à la e-cig. In vitro, l’exposition d’un modèle de barrière alvéolo-capillaire aux principaux composants des e-L a montré des effets néfastes sur l’intégrité de la barrière, l’inflammation, le stress oxydant et la mort cellulaire, pour les principaux composants des e-L et leurs mélanges. La présence d’arômes mentholé et fruité, ainsi que de nicotine dans la composition des e-L semble empirer les observations.</dcterms:abstract>
<dcterms:abstract xml:lang="en">E-cigarettes (e-cig) have not been proven to be effective and tolerable, in part because of a wide range of liquids (eL) containing vehicles, flavors, and nicotine at different concentrations, making scientific studies more complex. The work presented here concerns 3 levels of analysis. In humans, the results of the VAPexp'AIR clinical trial are the first to show that the formulation of the eL influences the mass of inhaled nicotine and its pharmacokinetic parameters. In mice, a post-smoking COPD model has been developed with pathological features such as immune cell recruitment in the lower airways associated with pulmonary emphysema. This model will be used as a control during exposure to e-cig. In vitro, exposure of an alveolar-capillary barrier model to the main components of e-L showed deleterious effects on barrier integrity, inflammation, oxidative stress and cell death, for the main components of e-L and their mixtures. The presence of minty and fruity flavors, as well as nicotine in the composition of e-L seems to worsen the observations.</dcterms:abstract>
