<dcterms:abstract xml:lang="fr">La vigne (Vitis) est sujette à de très nombreuses viroses causant des symptômes qui, en touchant à la vitalité de la plante ou aux fruits produits impactent lourdement la viticulture mondiale. Ces maladies sont causées notamment par le Grapevine Fanleaf Virus (GFLV), l’Arabis Mosaic Virus, le Grapevine Leafroll-associated Virus 3 (GLRaV-3) ou encore le Grapevine Pinot Gris Virus (GPGV). Pour la vaste majorité d’entre eux, il n’existe pas de résistance naturelle dans le matériel génétique. Toutefois, les Nanobodies (Nb), des petits peptides dérivés des parties variables des chaînes lourdes d’immunoglobuline sont capable d’induire de la résistance in planta contre le GFLV. Toutefois celle-ci demeure contournables par des variants du phytovirus. Afin d’améliorer cette résistance et de l’étendre à d’autres cibles, j’ai caractérisé des collections de Nbs dirigés contre le GFLV et l’ArMV afin de trouver des Nbs avec des propriétés intéressantes pour l’expression in planta. En fusionnant divers Nbs dirigés contre le GFLV et l’ArMV, j’ai aussi pu pyramider et diversifier la résistance tout en limitant le nombre de transgènes insérés. Enfin, j’ai mis au point le FLOP-YD pour FLuOrescent-Protein based Yeast Display, un nouveau système qui permet de sélectionner des Nbs contre des phytovirus difficiles à produire. Ainsi j’ai pu isoler et caractériser des Nbs dirigés contre le GLRaV-3 et le GPGV.</dcterms:abstract>
<dcterms:abstract xml:lang="en">Grapevine (Vitis) is exposed to numerous viruses causing symptoms that, by affecting the vitality of the plant or the fruits produced, have a heavy impact on viticulture worldwide. These diseases are caused by Grapevine Fanleaf Virus (GFLV), Arabis Mosaic Virus, Grapevine Leafroll-associated Virus 3 (GLRaV-3) or Grapevine Pinot Gris Virus (GPGV). For the vast majority of them, there is no natural resistance in grapevine germplasm. However, Nanobodies (Nb), small peptides derived from the variable parts of immunoglobulin heavy chains, can mediate in planta resistance against GFLV. However, this resistance can be bypassed by variants of the phytovirus.To improve this resistance and to extend it to other targets, I characterized collections of Nbs directed against GFLV and ArMV to find Nbs with interesting properties for better in planta expression and affinity for GFLV. By fusing various Nbs directed against GFLV and ArMV, I also managed to pyramid and diversify resistance while limiting the number of inserted transgenes. Finally, I developed FLOP-YD for FLuOrescent -Protein based Yeast Display, a new, simple Yeast Surface Display system to select Nbs against hard to produce phytoviruses. As a result, I isolated and characterized several Nbs directed against GLRaV-3 and GPGV.</dcterms:abstract>
