<dcterms:abstract xml:lang="fr">Dans un contexte radiothérapeutique, la majorité des dégâts d'un point de vue moléculaire découlent des effets dits indirects. Ces effets proviennent de la radiolyse de l'eau, qui se décompose sous l'influence des rayonnements pour former des espèces réactives envers les biomolécules. En raison de leur proportion importante dans les cellules, les protéines sont des cibles de choix pour ces effets. Cette thèse s'inscrit alors dans une étude fondamentale des mécanismes de radiolyse de biomolécules protéiques selon différents paramètres : le transfert d'énergie linéique (TEL) et le débit de dose. Le premier fait écho à l'hadronthérapie et le second à la thérapie FLASH.De la radiolyse de l'eau aux acides aminés, ces deux effets seront étudiés et comparés pour des rayonnements ionisants de différentes natures (photons, électrons, ions). Les ions étant capables de fragmenter lors de leur pénétration de la matière, cette propriété sera également étudiée à travers nos systèmes. Enfin, les compétences acquises sur la radiolyse de biomolécules protéiques seront appliquées pour le développement de films de bioplastiques à base de protéines sous irradiations sans ajout d'additifs ou de traitements ultérieurs.</dcterms:abstract>
<dcterms:abstract xml:lang="en">In a radiotherapeutic context, the majority of damage from a molecular point of view derives from so-called indirect effects. These effects arise from the radiolysis of water, which breaks down under the influence of radiation to form species reactive towards biomolecules. Because of their high proportion in cells, proteins are prime targets for these effects. This thesis is part of a fundamental study of the radiolysis mechanisms of protein biomolecules according to different parameters : linear energy transfer (LET) and dose rate. The former echoes particle therapy, the latter FLASH radiotherapy. From radiolysis of water to amino acids, these two effects will be studied and compared for different types of ionizing radiation (photons, electrons, ions). As ions are capable of fragmenting as they penetrate matter, this property will also be studied using our chemical systems. Finally, the skills acquired in the radiolysis of protein biomolecules will be applied to the development of protein-based bioplastic films under irradiation without the addition of additives or further processing.</dcterms:abstract>
