<dcterms:abstract xml:lang="fr">La douleur chronique est un problème de santé publique majeur et les analgésiques opioïdes présentent de nombreux effets secondaires et sont connus pour induire de la tolérance et de l’addiction. Il y a donc un besoin urgent de trouver des traitements alternatifs. GalR2 est un RCPG connu pour être impliqué dans la modulation de la douleur, toutefois son rôle précis n’est pas encore clair. Afin de permettre un contrôle spatio-temporel précis de ce récepteur nous avons utilisé la photopharmacologie pour développer les premiers dérivés photoactivables de la spexine, un neuropeptide endogène qui active spécifiquement GalR2. Nous avons synthétisé des dérivés photoisomérisables portant un motif azobenzène qui ont montré différents profils d’activité vis-à-vis de GalR2. A côté de cela, les dérivés photocage de la spexine comprenant une tyrosine protégée avec un o-NB ou une DEACM ont montré une photoactivation significative ce qui a amené au développement de la première sonde photoactivable de GalR2 qui pourra être utilisée sur l’animal vivant.</dcterms:abstract>
<dcterms:abstract xml:lang="en">Chronic pain is a major public health issue and opioid analgesics present many side effects and they are known to prone tolerance and addiction. So there is an urgent need for alternative treatments. GalR2 is a GPCR know to be involved in pain modulation, however its precise role is still unclear. To allow a precise spatio-temporal control of this receptor, we used photopharmacology to develop the first photoactivatable derivatives of the spexin, an endogenous neuropeptide that specifically activate GalR2. We synthesized photoswitchable derivatives bearing an azobenzene moiety and they demonstrated different activity patterns towards GalR2. On the other hand, photocage spexin derivatives bearing a tyrosine protected with o-NB or DEACM showed significant photoactivation leading to the development of the first photoactivatable probe for GalR2 which could be further use in living animal.</dcterms:abstract>
