<dcterms:abstract xml:lang="fr">Ces travaux de thèse ont porté sur la synthèse et la caractérisation de nouvelles sondes fluorescentes photo-modulables pour des applications en bioimagerie. Un nouveau mécanisme de photo-conversion a été décrit, la Conversion Induite par Photo-oxydation Dirigée (DPIC en Anglais), basé sur l’incorporation d’un groupement aromatique sensible à l’oxygène singulet (ASORM en Anglais) conjugué à un fluorophore. Sous irradiation, le fluorophore génère de l’oxygène singulet, oxydant l’ASORM et entraînant une rupture de la conjugaison, menant donc à un effet hypsochrome du fluorophore. Ce mécanisme a été appliqué et caractérisé à deux familles de fluorophores, les coumarines et les BODIPYs, menant à l’obtention de sondes présentant une photo-conversion du proche infrarouge vers le rouge, du rouge vers le vert et du vert vers le bleu. Ces sondes ont également pu être fonctionnalisées par des groupes ciblants permettant l’application du DPIC au sein de différentes organelles comme la membrane plasmique, les mitochondries, les gouttelettes lipidiques, mais également sur des protéines à l’aide de ligand HaloTag, élargissant ainsi la palette de sondes moléculaires photo-convertibles.</dcterms:abstract>
<dcterms:abstract xml:lang="en">During this thesis, the synthesis and the characterization of new fluorescent photoconvertible probes for bioimaging were carried out. A new mechanism of photoconversion was described, the Directed Photooxydation Induced Conversion (DPIC). This mechanism is based on the conjugation of an Aromatic Singlet Oxygen Reactive Moiety (ASORM) with a fluorophore. Under irradiation, the fluorophore generates singlet oxygen, oxidizing the sensitive aromatic leading to a disruption of the conjugation and thus to a hypsochromic shift of the fluorophore. This mechanism was applied and characterized on two families of fluorophores, the coumarins and BODIPYs, enabling to obtain several probes displaying photo-conversion probes from near infrared to red, red to green and green to blue. Those probes were functionalized with targeting moieties allowing their use in various organelles such as plasma membrane, mitochondria, lipid droplets as well as proteins thanks to a HaloTag ligand expanding the diversity and versatility of molecular photoconvertible probes.</dcterms:abstract>
