<dcterms:abstract xml:lang="fr">La voie orale est la voie d’administration privilégiée pour administrer les médicaments. Les barrières liées à l’absorption de principes actifs par voie orale ont poussé les formulateurs à employer plusieurs stratégies pour améliorer l’absorption des principes actifs, par exemple, en ayant recours à des systèmes d’encapsulation tels que les nanoémulsions. Les nanoémulsions permettent d’encapsuler les molécules peu solubles et de les rendre aptes à traverser les différentes barrières membranaires. Elles permettent de les véhiculer au site d’absorption et/ou d’action. Trois axes ont été développés à travers cette thèse. Le premier axe consiste à préparer des nanoémulsions inverses (E/H) en utilisant l’ultrasonication. Plusieurs stratégies ont été adoptées afin d’essayer de produire des nanoémulsions inverses dont les gouttelettes sont les plus fines et les plus homogènes possible. Le deuxième axe porte sur la préparation de lyophilisats oraux qui libèrent une nanoémulsion. La nanoémulsion libérée est une nanoémulsion directe (H/E). Le troisième axe propose une mise en forme plus avancée. Il s’agit de préparer des lyophilisats innovants afin d’obtenir une libération plus rapide de la nanoémulsion.</dcterms:abstract>
<dcterms:abstract xml:lang="en">Oral route is the most used route to administer the medicaments. Barriers related to the absorption of active ingredients pushed the formulators to employ multiples strategies to enhance the absorption of active ingredients, for example, by using encapsulation systems like nanoemulsions. Nanoemulsions can encapsulate poorly soluble molecules and make them able to go through different membranes barriers. They can deliver them to the site of absorption and/or action. Three axes have been developed through this thesis. The first one is based on the preparation of reverse nanoemulsions (W/O) by using ultrasonication. Multiple strategies have been adopted to have the finest and the most homogenous droplets. The second axe is based on the preparation of oral lyophilizates that release nanoemulsion. The released nanoemulsion is a direct nanoemulsion (O/W). The third axe proposes an advanced pharmaceutical form. It aims to prepare innovative lyophilizates to have faster release of nanoemulsion.</dcterms:abstract>
