<dcterms:abstract xml:lang="fr">La vitamine A est une petite molécule ayant un nombre très important de fonctions dans l’organisme et, de fait, une implication dans une large variété de pathologies. Aujourd’hui, la connaissance de son métabolisme est incomplète, en particulier à l’échelle cellulaire, ce qui se répercute sur la compréhension de pathologies complexes telles que la maladie de Parkinson. Ainsi, le but de cette thèse est de développer de nouveaux outils moléculaires permettant de localiser les rétinoïdes aux échelles cellulaire et subcellulaire mais également de détecter et d’identifier de nouveaux métabolites. Ces études fourniront les bases qui contribueront à élargir la compréhension du métabolisme et du stockage des rétinoïdes. La première partie de cette thèse décrit la mise au point d’une nouvelle méthode d’insertion tardive d’azoture sur la vitamine A afin d’en faire une sonde moléculaire détectable. La seconde partie présente la conception et la synthèse de sondes fluorescentes pour leur application en imagerie cellulaire dans un but de visualisation des rétinoïdes aux échelles cellulaire et subcellulaire. Enfin, la dernière partie retrace le développement de sondes de spectrométrie de masse facilitant la détection et la quantification de nouveaux métabolites de la vitamine A.</dcterms:abstract>
<dcterms:abstract xml:lang="en">Vitamin A is a small molecule with many functions in the body and is therefore involved in a wide range of pathologies. Today, our knowledge of its metabolism is incomplete, particularly at the cellular level, which has implications for our understanding of complex pathologies such as Parkinson's disease. The aim of this thesis is therefore to develop new molecular tools to localize retinoids at the cellular and subcellular levels and to detect and identify new metabolites. These studies will provide the basis for a broader understanding of retinoids’ metabolism and storage. The first part of this thesis describes the development of a new method of late stage azidation of vitamin A to make it a detectable molecular probe. The second part presents the design and synthesis of fluorescent probes for their application in cellular imaging to visualize retinoids at cellular and subcellular levels. Finally, the last part describes the development of mass spectrometry probes to facilitate the detection and quantification of new vitamin A metabolites.</dcterms:abstract>
