<dcterms:abstract xml:lang="fr">Le transfert d'énergie par résonance sombre (DRET) est un mécanisme de transfert d'énergie utilisant un donneur "sombre". L’objet de cette thèse était d'étudier les caractéristiques photophysiques d'un système DRET intermoléculaire utilisant un substitut de nucléobase DiethylaminoFluroreneKetotriazolyl (X) comme donneur "sombre" et de l'appliquer à l'imagerie de super-résolution DNA-PAINT. La nature fluorogène et le grand « Stokes shift » de X sont une avancée par rapport aux colorants Förster-RET. L'évolution de la DRET en fonction de la distance entre X et l'accepteur Cy5 dans les duplex d'ADN montre une nature oscillante corrélée à l'orientation relative entre X et Cy5. La dépolarisation induite par le transfert d'énergie mesurée par l'anisotropie montre une nature oscillante similaire. La mise en œuvre du DRET pour la microscopie de super-résolution DNA-PAINT a montré un fort potentiel de cette approche en obtenant des images de super-résolution de microtubules cellulaires en moins de 30 s.</dcterms:abstract>
<dcterms:abstract xml:lang="en">Dark resonance energy transfer (DRET) is an energy transfer mechanism using a quenched "dark" donor. The aim of this thesis was to study the photophysical characteristics of an intermolecular DRET system using the nucleobase substitute DiethylaminoFluroreneKetotriazolyl (X) as a "dark" donor, and to apply it to DNA-PAINT super-resolution imaging. The fluorogenic nature and high "Stokes shift" of X represent an advancement over Förster-RET dyes.The evolution of DRET as a function of the distance between X and the Cy5 acceptor in DNA duplexes shows an oscillating nature correlated to the relative orientation between X and Cy5. The energy transfer induced depolarization measured by anisotropy shows a similar oscillating nature.The implementation of DRET on the DNA-PAINT super-resolution microscopy demonstrated the great potential of this new approach, obtaining super-resolution images of cellular microtubules in less than 30 s.</dcterms:abstract>
