<dcterms:abstract xml:lang="fr">Les cancers hautement agressifs, comme les cancers du sein triple négatifs (TNBC) et les glioblastomes, restent à l'heure actuelle fatals dans la plupart des cas à cause du manque de traitements curatifs. Cet échec thérapeutique peut être, entre autres, attribué à la survenue de mécanismes de résistance aux thérapies conventionnelles, mais aussi à l'hétérogénéité des tumeurs ainsi qu'à la présence de sous-populations de cellules souches cancéreuses (CSC), souvent responsables des rechutes. Ce besoin médical non satisfait a rendu nécessaire l'exploration de nouvelles cibles thérapeutiques jouant un rôle important dans ce type de cancers. L'enzyme poly(ADP-ribose polymérase 3 (PARP3) a été récemment décrite comme un promoteur de l’agressivité tumorale et du maintien des propriétés souches des cellules. Ces différents éléments renforcent l'intérêt de l'étude de PARP3 dans un contexte de thérapie contre des cancers agressifs, mais aussi de son rôle dans les CSC. Au cours de cette thèse, nous avons donc mis en évidence la pertinence du ciblage de PARP3 dans les cellules de TNBC et les glioblastomes reposant sur une thérapie combinée faisant intervenir un petit ARN, interférant avec l'expression de PARP3 (siPARP3), et un composé dont les effets sont potentialisés lorsque PARP3 est déplétée. Nous avons par la suite mis au point des stratégies de vectorisation de ces molécules par une approche de conjugués anticorps-médicaments (ADC) en utilisant un anticorps modèle, le trastuzumab (anti-HER2), pour réaliser la preuve de concept de notre stratégie. Nous avons alors pu valider l’efficacité de ces conjugués in vitro dans des lignées cancéreuses HER2+. En parallèle, nous avons mené une étude du rôle de PARP3 dans les cellules souches de glioblastome (GSC) dérivées de patients dans laquelle PARP3 a été disruptée par la technique CRISPR/Cas9. Nous avons ainsi montré qu’une déficience en PARP3 entrainait une perte des propriétés souches de ces cellules, telles que la clonalité, l’autorenouvellement ou encore leur capacité à former des tubes vasculogéniques.</dcterms:abstract>
<dcterms:abstract xml:lang="en">Highly aggressive cancers, such as triple-negative breast cancer (TNBC) and glioblastoma, are in most cases fatal due to the lack of curative treatments. This therapeutic failure can be attributed, among other factors, to the emergence of resistance mechanisms to conventional therapies, as well as to tumor heterogeneity and the presence within tumors of subpopulations of cancer stem cells (CSC) often responsible for relapses. This unmet medical need has necessitated the exploration of new therapeutic targets that play an important role in these types of cancers. The poly(ADP-ribose) polymerase 3 (PARP3) enzyme has recently been described to promote tumor aggressiveness as well as the maintenance of stem cell properties. These various elements reinforce the interest in studying PARP3 in the context of therapy against aggressive cancers, as well as its role in CSCs. In this thesis, we have highlighted the relevance of targeting PARP3 in TNBC and glioblastoma cells, using a combined therapy approach involving small interfering RNAs targeting PARP3 expression (siPARP3) and a compound whose effects are potentiated when PARP3 is depleted. Subsequently, we developed vectorization strategies for these molecules using an antibody-drug conjugate (ADC) approach on a model antibody, trastuzumab (anti-HER2), to perform the proof of concept of our strategy. We then validated the efficacy of these conjugates, in vitro, in HER2+ cancer cell lines. In parallel, we conducted a study on the role of PARP3 in patient-derived glioblastoma stem cells (GSC), in which PARP3 was disrupted using the CRISPR/Cas9 technique. We showed that PARP3 deficiency leads to a loss of stem cell properties in these cells, including clonality, self-renewal as well as their ability to form vasculogenic tubes.</dcterms:abstract>
