<dcterms:abstract xml:lang="fr">Les carcinomes épidermoïdes de la tête et du cou (CETEC) sont des cancers hétérogènes diagnostiqués tardivement. L’absence de biomarqueurs diagnostiques, pronostiques de la survie des patients ou prédictifs de la réponse au traitement est un problème majeur, en lien avec la survenue fréquente de rechutes ou de métastases et d’une survie faible des patients. Dans ce projet de thèse, nous avons identifié les miR-30a-3p et miR-30e-3p comme biomarqueurs pronostiques de la survie et des rechutes locorégionales ou métastatiques des patients CETEC. Nous avons validé leur rôle de suppresseurs de tumeur au travers du ciblage de membres de la superfamille du TGF-β (TGFBR1 et BMPR2). Enfin, nous avons mis en évidence le rôle immunomodulateur des miR-30a/e-3p, qui influencent l’expression de cytokines chemoattractantes et modifient le comportement des cellules immunitaires telles que les macrophages et les lymphocytes autour de la tumeur, tout en augmentant l’expression de PD-L1 sur les cellules tumorales et PD-1/PD-L1 sur les macrophages.</dcterms:abstract>
<dcterms:abstract xml:lang="en">Head and neck squamous cell carcinomas (HNSCC) are heterogeneous tumors frequently diagnosed at an advanced stage. The lack of diagnostic, prognostic, and predictive biomarkers is a major issue, contributing to frequent relapses, metastases, and low survival rates. In this project, we have identified miR-30a-3p and miR-30e-3p as prognostic biomarkers for the survival and locoregional relapses or metastatis of HNSCC patients. We validated their role as tumor suppressors through the targeting of TGF-β superfamily members (TGFBR1 and BMPR2). Furthermore, we highlighted the immunomodulatory role of miR-30a/e-3p as they influence the expression of chemoattractive cytokines, alter the behaviour of immune cells such as macrophages and lymphocytes around the tumor and increase the expression of PD-L1 on tumor cells and PD-1/PD-L1 on macrophages.</dcterms:abstract>
