<dcterms:abstract xml:lang="fr">Le but de cette thèse est d’explorer les relations structure/propriétés de protéines de novo capables de catalyser efficacement le transfert de groupes acyles, notamment la formation de liaisons amide entre deux peptides par NCL en conditions natives et à des concentrations sub-milimolaires. En utilisant différents types de thiols comme résidus catalytiques, nous avons démontré la capacité de modifier les propriété de ces catalyseurs. Nous avons introduit un second site catalytique et avons tenté d’orienter le mécanisme d’un fonctionnement compétitif à un fonctionnement coopératif. Les résultats obtenus représentent une avancée significative vers le développement de futurs catalyseurs protéiques pour la ligation et la cyclisation de peptides.</dcterms:abstract>
<dcterms:abstract xml:lang="en">The goal of this thesis is to explore the structure/property relationships of de novo proteins capable of efficiently catalyzing the transfer of acyl groups, particularly the formation of amide bonds between two peptides through Native Chemical Ligation (NCL) under native conditions and at sub-millimolar concentrations. By using different types of thiols as catalytic residues, we demonstrated the ability to modify the properties of these catalysts. We introduced a second catalytic site and attempted to shift the mechanism from a competitive to a cooperative function. The results obtained represent a significant advancement toward the development of future protein catalysts for peptide ligation and cyclization.</dcterms:abstract>
