<dcterms:abstract xml:lang="fr">Les aminoglycosides sont des antibiotiques à large spectre qui se caractérisent par leur bactéricidie rapide et par un effet post-antibiotique important. Suite à leur découverte, les aminoglycosides ont servi à traiter des maladies infectieuses ayant ravagé l’humanité depuis la nuit des temps. Toutefois, malgré leur activité intrinsèque, la toxicité de ces antibiotiques, ainsi que l’apparition de souches bactériennes résistantes dotées d’enzymes modifiant les aminoglycosides (AMEs) a dramatiquement diminué leur domaine d’application. Cela engendre un besoin croissant pour le développement d’aminoglycosides capables de contourner ces mécanismes. Dans ce contexte, ces travaux de thèse se focalisent sur la synthèse de nouveaux aminoglycosides capables de surmonter la désactivation enzymatique catalysée par les AMEs. Ainsi, la modification ciblée de la position 5’’ de la néomycine B a permis la synthèse d’une première génération de dérivés invulnérables à l’enzyme ANT(4’)-Ia de S. aureus. En couplant ce type de modification avec une manipulation sélective de la position 3’, nous avons réussi à concevoir un aminoglycoside réfractaire aux AMEs de type ANT(4’)-Ia et APH(3’)-IIIa de S. aureus. La modification ponctuelle de l’amikacine nous a conduits vers des dérivés hémisynthétiques échappant simultanément à l’ANT(4’)-Ia et à l’APH-(3’)-IIIa sur des souches résistantes de S. aureus tout en préservant un bon profil antibiotique sur des isolats cliniques résistants de E. coli.</dcterms:abstract>
<dcterms:abstract xml:lang="en">Aminoglycosides are a class of broad-spectrum antibiotics characterized by their rapid bactericidal action and significant post-antibiotic effect. Following their discovery, aminoglycosides were used to treat infectious diseases that had ravaged mankind since the dawn of time. However, despite their intrinsic activity, the toxicity of these antibiotics, as well as the appearance of resistant bacterial strains bearing aminoglycoside-modifying enzymes (AMEs), has dramatically reduced their field of application. This phenomenon led to a dire need for the development of new antibiotics capable of bypassing these mechanisms.In this context, this thesis focuses on the synthesis of new aminoglycosides capable of overcoming enzymatic deactivation catalyzed by AMEs. Targeted modifications of neomycin B enabled us to synthesize a first generation of derivatives invulnerable to the ANT(4‘)-Ia enzyme of S. aureus. By combining this modification with selective manipulation of the 3’ position, we succeeded in designing an aminoglycoside invulnerable to ANT(4‘)-Ia and APH(3’)-IIIa AMEs from S. aureus. Selective modification of amikacin led to a series of compounds capable of simultaneously escaping ANT(4‘)-Ia and APH-(3’)-IIIa enzymes in resistant strains of S. aureus while exhibiting an interesting antibiotic profile against resistant clinical isolates of E. coli.</dcterms:abstract>
