<dcterms:abstract xml:lang="fr">La microfluidique en gouttelettes est une technologie permettant de fortement miniaturiser les réactions chimiques et biologiques en les réalisant au sein de compartiments de quelques picolitres très homogènes en taille. Cette technologie repose sur la génération de gouttelettes à partir de deux phases non miscibles, une phase aqueuse et une huile, manipulées dans des micropuces composées de canaux de quelques micromètres de large. De plus, la stabilité des gouttelettes est assurée par l’ajout d’un tensioactif qui se place à l’interface eau/huile et n’interagit habituellement pas avec le contenu de la gouttelette. Les travaux menés dans le cadre de cette thèse visent principalement à développer une nouvelle classe de tensioactifs fonctionnalisés, facilitant le greffage ciblé de biomolécules sur la surface des gouttes produites par microfluidique. Cette démarche s'appuie sur une chimie click, en particulier la réaction de SPAAC (cycloaddition azoture-alcyne tendu). L'approche adoptée repose sur l'utilisation de tensioactifs fluorés, modifiés par des groupements azotures, qui peuvent interagir avec des alcynes tendus présents sur des biomolécules telles que les anticorps ou les acides nucléiques, permettant leur fixation à la surface interne des gouttelettes. Ainsi, ces nouvelles molécules permettent d’exploiter la surface interne des gouttelettes et ouvrent de nouvelles perspectives applicatives à la technologie.</dcterms:abstract>
<dcterms:abstract xml:lang="en">Droplet microfluidics is a technology that makes it possible to greatly miniaturize chemical and biological reactions by carrying them out in compartments of a few picolitres that are very homogeneous in size. This technology is based on the generation of droplets from two immiscible phases, an aqueous phase and an oil, manipulated in microchips composed of channels a few micrometers wide. In addition, droplet stability is ensured by the addition of a surfactant that is placed at the water/oil interface and does not usually interact with the contents of the droplet.The main aim of the work carried out in this thesis is to develop a new class of functionalized surfactants, facilitating the targeted grafting of biomolecules onto the surface of droplets produced by microfluidics. This approach is based on click chemistry, in particular, the SPAAC (strained azide- alkyne cycloaddition) reaction. The approach adopted is based on the use of fluorinated surfactants, modified by azide groups, which can interact with strained alkynes present on biomolecules such as antibodies or nucleic acids, enabling them to be attached to the internal surface of droplets. These new molecules make it possible to exploit the inner surface of droplets, opening up new applications for the technology.</dcterms:abstract>
