<dcterms:abstract xml:lang="fr">Avec leur rôle clef dans les mécanismes physiopathologiques auto-immuns et tumoraux, les cellules immunitaires sont des cibles thérapeutiques particulièrement intéressantes à, selon le contexte, éliminer ou activer spécifiquement pour rétablir des fonctions immunitaires perturbées. Pour de telles approches thérapeutiques, les nanoparticules, grâce à leur versatilité, suscitent un grand intérêt. En effet, elles peuvent, grâce à l’ajout de motifs ou molécules actives, pendant ou après leur synthèse, devenir de véritables plateformes multifonctionnelles. Dans ce contexte, l’objectif de mes travaux de thèse a été de développer deux types de nanoparticules à visée thérapeutique dont les effets reposent sur le ciblage spécifique de cellules immunitaires. D’une part, dans le contexte de la polyarthrite rhumatoïde, des nanotubes de carbone fonctionnalisés ont été évalués pour une application de thérapie photothermique ablative ciblant les macrophages synoviaux. D’autre part des pseudo-particules virales décorées et adressées aux cellules dendritiques ont été développées comme plateformes de vaccination anti-tumorale.</dcterms:abstract>
<dcterms:abstract xml:lang="en">Given their key role in autoimmune and tumor-related pathophysiological mechanisms, immune cells are particularly promising therapeutic targets. Depending on the context, to restore disrupted immune functions, they can be either specifically eliminated or activated. For these therapeutic approaches, nanoparticles, due to their high versatility, are of great interest. Indeed, with the addition of targeting agents or active molecules during or after their synthesis, nanoparticles can become multifunctional structures. In this context, my thesis project aimed at developing two types of therapeutic nanoparticles whose effects rely on the specific targeting of immune cells. On one hand, in the context of rheumatoid arthritis, functionalized carbon nanotubes were evaluated for photothermal therapy to deplete synovial macrophages. On the other hand, antigen-containing dendritic cell-targeted viral-like particles were developed as cancer vaccine platforms.</dcterms:abstract>
