<dcterms:abstract xml:lang="fr">Les aptamères d'ARN sont des acides nucléiques monocaténaires capables d'adopter diverses conformations pour reconnaître spécifiquement une large gamme de ligands, y compris de petites molécules et des protéines. En plus de leur capacité de reconnaissance, ces aptamères peuvent exercer des fonctions complexes, ce qui les rend prometteurs en tant qu'outils thérapeutiques et diagnostiques. Ils trouvent des applications variées, allant de l'inhibition d'enzymes à la détection précise de petites molécules dans différents contextes. Dans cette thèse, la combinaison des méthodes SELEX et μIVC-seq a permis ’ x différentes fonctions. Elle a conduit à la sélection de biosenseurs de petites molécules avec des applications en détection environnementale, ainsi qu'au développement d'aptamères inhibiteurs d'enzymes, stables et efficaces dans des milieux complexes.</dcterms:abstract>
<dcterms:abstract xml:lang="en">RNA aptamers are single-stranded nucleic acids capable of adopting various conformations to specifically recognize a wide range of ligands, including small molecules and proteins. Beyond their recognition ability, these aptamers can perform complex functions, making them promising as therapeutic and diagnostic tools. They have diverse applications, ranging from the inhibition of enzymes to the precise detection of small molecules in different contexts. In this thesis, the combination of SELEX and μIVC-seq methods has been leveraged to exploit these various functions. This approach led to the selection of small molecule biosensors with environmental detection applications, as well as the development of enzyme-inhibiting aptamers that are stable and effective in complex environments.</dcterms:abstract>
