<dcterms:abstract xml:lang="fr">Dans le cytoplasme le précurseur Gag du VIH-1, sélectionne l’ARN génomique (ARNg) viral en interagissant spécifiquement avec sa région 5' UTR. Ensuite, Gag permet l’ancrage de l’ARNg aux sites d’assemblage à la membrane plasmique (MP). A ces sites, pour construire la particule virale, environ 2500 copies de Gag oligomérisent sur l’ensemble de l’ARNg . Cela suggère qu’à ce stade les interactions entre Gag et la 5’UTR de l’ARNg doivent devenir non spécifiques. Lors de ma thèse j’ai cherché à caractériser le rôle des facteurs moléculaires tels que l’association de Gag à la MP et sa multimérisation aux sites d’assemblage, sur la spécificité des interactions Gag – 5’UTR de l’ARNg. Cette étude, conduite par microcopie confocale dans un système in vitro imitant les sites d’assemblage du VIH-1, a permis de mettre en évidence que i) l’association de Gag à la membrane induit des changements dans le mode de liaison Gag – 5’UTR de l’ARNg, ii) l’interface de symétrie triple sur le réseau d’assemblage de la capside est requise pour l’interaction à forte affinité entre Gag et la 5’UTR et iii) la protéine Gag s’assemble de manière plus efficace à la membrane lorsqu’elle interagit avec la 5’UTR de l’ARNg plutôt qu’un autre ARN. En parallèle, pour caractériser les interactions Gag–ARNg à la membrane à l’échelle atomique, j’ai mis en place des stratégies permettant de former des complexes lipides-Gag et lipides-Gag-5’UTR de l’ARNg sur des grilles de cryo-microscopie électronique fonctionnalisées avec des monocouches lipidiques ou des liposomes.</dcterms:abstract>
<dcterms:abstract xml:lang="en">In the cytoplasm, the HIV-1 precursor Gag selects viral genomic RNA (gRNA) by interacting specifically with its 5' UTR region. Then, Gag allows the anchoring of the gRNA to the assembly sites on the plasma membrane (PM). At these sites, to build the viral particle, around 2.500 copies of Gag oligomerize onto the entire gRNA. This suggests that, at this stage, the interactions between Gag and the 5'UTR of the gRNA must become non-specific. During my thesis, I sought to characterize the molecular factors such as the association of Gag with the PM and its multimerization at assembly sites, on the specificity of Gag - 5'UTR gRNA interactions. This study, conducted by confocal microcopy in an in vitro system mimicking HIV-1 assembly sites, revealed that i) the association of Gag with the membrane induces changes in the Gag - 5'UTR binding mode, ii) the 3-fold symmetry interface on the capsid assembly lattice is required for the high-affinity interaction between Gag and the 5'UTR and iii) the Gag protein assembles more efficiently at the membrane when it interacts with the 5'UTR of the gRNA rather than another RNA. In the meantime, to characterize the Gag-gRNA interactions at the membrane at the atomic scale, I set up strategies to form lipid-Gag and lipid-Gag-5'UTR complexes on cryo-electron microscopy grids, functionalized with lipid monolayers or liposomes.</dcterms:abstract>
