<dcterms:abstract xml:lang="fr">La microscopie de fluorescence est couramment utilisée en biologie car elle permet d’imager en volumes avec contraste et spécificité. L’utilisation d’une illumination en feuille de lumière réduit la phototoxicité et le photoblanchiment, mais complexifie le montage des échantillons. Un de ses dérivés, la microscopie à plan oblique (OPM), permet de monter les échantillons de manière classique, sur lame ou via des plaques multi-puits. Un intérêt a, ici, été porté à développer et améliorer cette technique.Premièrement, les OPM nécessitent des lentilles de tube de focale précise, non disponibles dans le commerce. Un logiciel en source ouverte a été développé pour en générer en limitant les aberrations, à partir de deux lentilles achromatiques. Secondement, un OPM a été conçu et monté. Son immersion à huile réduit le coût tout en augmentant l’ouverture numérique, par rapport à une immersion à eau. Des acquisitions de C. elegans adultes et de larves de poissons zèbres ont été réalisées.</dcterms:abstract>
<dcterms:abstract xml:lang="en">Biologists are used to fluorescence microscopy, as it allows to obtain volumetric images with contrast and specificity. Lightsheet illumination reduces phototoxicity and photobleaching, but complicates sample mounting. Oblique Plane Microscopy (OPM), which stems from it, allows the use of conventional mounting, such as microscope slides or multiwell plates. This research aimed at developing and improving this technique.Firstly, OPM needs tube lenses with specific focal lengths, often not commercially available. An open-source software was developed to design them by combining two achromatic lenses. Optical aberrations are considered. Secondly, an OPM was devised and assembled. It uses oil immersion, instead of water, in order to improve the numerical aperture while reducing the price. Adult C. elegans and Zebrafish larvae were imaged.</dcterms:abstract>
