<dcterms:abstract xml:lang="fr">Ce manuscrit présente les travaux réalisés pour développer et optimiser une méthode de concentration virale par ultrafiltration sur membrane, adaptée au traitement de grands volumes d’eaux usées, en réponse aux besoin croissant de l’épidémiologie basée sur les eaux usées. Dans un premier temps, la méthode a été évaluée en termes d’efficacité d’enrichissement, de sensibilité, de répétabilité et de compatibilité avec le séquençage. Elle a permis une quantification fiable et sensible par RT-qPCR et RT-dPCR, ainsi qu’un séquençage de haute qualité du génome complet du SARS-CoV-2, avec une couverture dépassant 90 %. Les rendements de concentration varient selon les échantillons et le type de virus analysé. Une étude complémentaire a donc été menée afin d’optimiser cette méthode en explorant le comportement des virus dans les eaux usées, notamment leur distribution entre les fractions solide et liquide ainsi que l’influence des paramètres physico-chimiques sur les rendements de concentration.</dcterms:abstract>
<dcterms:abstract xml:lang="en">This manuscript presents the work conducted to develop and optimize a membrane-based ultrafiltration method for concentrating viruses from large volumes of wastewater, addressing the growing needs of wastewater-based epidemiology. The method was first evaluated for its enrichment efficiency, sensitivity, repeatability and suitability for sequencing. In enabled reliable quantification by RT-qPCR and RT-dPCR, as well as high-quality whole genome sequencing of SARS-CoV-2 with coverage exceeding 90%. Concentration yields varied depending on the sample and the virus type. Therefore, an additional study was carried out to optimize the method by investigating the behavior of viruses in wastewater, including their distribution between solid and liquid fractions, and the influence of physicochemical parameters on concentration efficiency.</dcterms:abstract>
